| Objective To explore the influence of modified Huangqijianzhongtang on expression of ki67,p53 in Lewis lung cancer model mice with spleen deficiency syndrome and the inhibitory effect of lung cancer cell proliferation and possible mechanism.Method Spleen Deficiency model mice was established by Composite factors methods,such as diarrhea caused by cold and bitter Traditional Chinese medicine、overstrain、intemperance of taking food.The mice was grouped according to the experiment design and observed at the same time.Mice’s eyeballs blood was tested for the serum D-xylose level;small intestinal mucosa was examed to confirm the success of model establishment.Then spleen deficiency Lewis lung cancer mouse model was established by inoculating tumor strain.Mice were killed after 11 days intragastric administration with modified Huangqijianzhongtang.Remove the tumor tissue and paraffin-embedded it.Immunohistochemistry was used to detect the expression of ki-67 and p53.TUNEL assay was used to detect the apoptosis of tumor cells.Result The Lewis lung cancer mice model with spleen deficiency was established.The general physical condition of mice with spleen deficiency was poor,the serum D-xylose level were significantly lower;the structure of intestinal mucosa was damaged obviously.The expression of ki-67,p53 of lung cancer spleen deficient group were higher than lung cancer group,AI was lower than lung cancer group.The expression of ki-67 of Modified HuangQiJianZhongTang group was higher than spleen deficient lung cancer group,p53 and AI was lower than spleen deficient lung cancer group.Conclusion The expression of ki-67 was up-regulated and the expression of p53 was decreased in lung cancer tissues of spleen deficiency syndrome,and AI was decreased.The proliferation rate of tumor cells was significantly higher than lung cancer group.The expression of ki-67 was decreased,the expression of p53 was up-regulated and the AI was increased in the modified Huangqijianzhongtang,tumor cell proliferation rate slowed down.The results indicated that modified HuangqiJianzhongtang could inhibit the proliferation of Lewis lung cancer cells in spleen deficiency syndrome by regulating the positive expression of p53 and ki67.The results indicated that modified Huangqij ianzhongtang could inhibit the proliferation of Lewis lung cancer cells in spleen deficiency syndrome by regulating the positive expression of p53 and ki-67. |
