Study On The Protective Effect Of Different Effective Parts Of Penthorum Chinense Pursh On Alcoholic Liver Injury In Mice

Objective:Modern research shows that Penthorum chinense Pursh containing active ingredients,has a protective effect on the liver,can reduce the damage of alcohol and drugs on the liver,but the effective part of the protective effect is still unclear.This study used alcohol induced liver injury mouse model,comparative study of Penthorum chinense Pursh with effective parts(ethanol,ethyl acetate,n-butanol,petroleum ether)protective effect on acute alcoholic liver injury in mice,to determine the location of Penthorum chinense Pursh in treatment of alcoholic liver injury,and to provide the experimental basis for the scientific basis and clinical application of Penthorum against alcoholic liver injury.Methods:1.Penthorum chinense Pursh preparation of different effective parts:respectively with 4 different organic solvents(ethanol,ethyl acetate,n-butanol,petroleum ether),on the same quality Penthorum chinense Pursh added quality is two times the mass of Penthorum chinense Pursh catch diatomite was soaking,heating reflux extraction,filtrate rotary evaporator,water bath concentration,volatile dry,vacuum drying.The extract rutin,by spectrophotometry(Chinese Pharmacopoeia 2015 VB)determination of total flavonoids in different Penthorum chinense Pursh in effective parts.2.Penthorum chinense Pursh screening study on the protective effect of alcohol induced acute liver injury of different fractions:using alcohol(56 degree liquor 20ml/kg/d)by gavage for alcohol induced liver injury in mice by 4 weeks and supplemented with high iron diet induced by Tiopronin,as positive control(50mg/kg/d),4 kinds of Penthorum chinense Pursh effective parts are set low,high dose group(5g.crude drugs herbs/kg/d、(10g.crude drugs herbs/kg/d).Intragastric administration for 4 weeks,transaminase detect Penthorum chinense Pursh fractions on alcoholic liver injury model of mouse serum alanine(ALS),aspartate aminotransferase(AST),three glycerol(TG),total cholesterol(TC),tumor necrosis factor alpha(TNF-alpha)superoxide dismutase and liver in the organization(SOD),malondialdehyde(MDA)level and liver macroscopic observation and histological examination.Results:1.successfully prepared Penthorum chinense Pursh 4 effective parts,namely ethanol fraction,ethyl acetate,n-butanol,petroleum ether extract,total flavonoids content in effective parts:2.34%ethanol parts:5.61%;ethyl acetate;butanol 2.13%;petroleum ether fraction:0.97%.2.4 weeks after the model mice were sacrificed at random,through the liver tissues were found in alcoholic liver injury in mice was successful.4 kinds of effective parts Penthorum chinense Pursh significantly reduce the alcoholic liver injury in mice serum AST,ALT,TC,TG level,decrease the content of MDA in liver and enhance the activity of SOD,reduce the levels of TNF-α in serum,and improve the pathological changes of alcoholic liver injury in mice,compared with the model group,the difference was statistically significant(P<0.05orP<0.01).comparison between the two groups Penthorum chinense Pursh the strongest protective effect on ethanol part of alcoholic liver injury in mice,and showed similar drug tiopronin group,and the protective effect of petroleum ether fraction is relatively poor.Conclusion:Penthorum chinense Pursh ethanol fraction,ethyl acetate,n-butanol,petroleum ether extract has protective effect on alcoholic liver injury in mice.The mechanism may be related to scavenging free radicals and anti lipid peroxidation and reduce the expression of serum inflammatory factor TNF-α.The protective effect of ethanol extract of Penthorum chinense Pursh catch the best,that may be related with high content of total flavonoids in the ethanol parts,parts can be used as the main effective part of Penthorum chinense Pursh against alcoholic liver injury.
