| Objective:Paraboea rufescens Franch.is the species of Paraboea genus of the family Gesneriaceae.It is also known as Yanwoju,Xiucao,Yanpipa.etc.As a folk herb it distributes mainly in Guizhou,Guangxi province.The whole plant of P.rufescens was used for the treatment of cough,labor injury,carbuncle sore,cold,hematemesis and so on.Previous studies have shown that the genus Gesneriaceae plants contain a large number of phenylethanoid glycosides,and such kind of components is closely related to the taxonomy of the Gesneriaceae plants.Therefore,in this study,the phenylethanol glycoside constituents were used as the target,a phytochemical study of P.rufescens was carried out,and furthermore chemical characterization of 32 medicinal plants of the genus Gesneriaceae were conducted with the aim to explore phylogenetic development of the plant group.Methods:1.Using conventional extraction and separation methods,solvent extraction,column chromatography and HPLC preparation to systematically isolate and purify the phenylethanol glycosides from P.rufescens.2.Using modern spectroscopic technology such as ultraviolet spectroscopy(UV),infrared spectroscopy(IR),high resolution electrospray ionization mass spectrometry(HR-ESI-MS)and nuclear magnetic resonance(1D-NMR,2D-NMR)to identify and elucidate the isolated compounds.3.UPLC-MS/MS method was applied to simultaneously determine phenylethanol glycosides in 32 common medicinal plants of Gesneriaceae.Results and conclusion:1.Penylethylethanol glycosides of P.rufescens27 compounds were isolated from the 50%methanol fraction of the macroporous resin extract,and the structures of 19 compounds were identified and elucidated,these compounds included nuomioside A(K1),isonuomioside A(K2),paraboside B(K6),paraboside A(K7),conandroside(K10),K11,K12,K13,K14,K16,K17,K18,K19,K21,K22,K23,K24,K26,K27,where K11,K19,K24,K12,K13,K14,K16,K17,K18,K21,K26,K22,K23,K27 are 14 new phenylethanol glycosides,and named as rufescenside Ⅰ(K11),rufescenside Ⅱ(K19)and rufescenside Ⅲ(K24),rufescenside A(K12),rufescenside B(K13),rufescenside C(K14),rufescenside D(K16),rufescenside E(K17),rufescenside F(K18),rufescenside G(K21)and rufescenside H(K26),rufescenside R(K22),rufescenside S(K23),rufescenside T(K27),respectively.All the ingredients were isolated from the wild P.rufescens for the first time.2.Determination of phenylethanoid glycosides in common medicinal plants of Gesneriaceae.Samples of 19 populations were collected from Hunan,Yunnan,Sichuan and Chongqing,and samples of 13 populations were collected from Guilin.The content of phenylethanoid glycosides of all samples were quantitative determined.The results showed that phenylethanol glycosides ingredients were widely found in the medicinal plants of the genus Gesneriaceae,but there existed some differences among the species. |
