Phenylethanoid glycosides (PhGs) (mainly contain echinacoside and acteoside) are major bioactive phenyethanoids in medicinal herba Cistanche tubulosa (Schenk) R. Wight and has been reported to have antioxidant and anti-aging in many experimental studies. In this work, phenylethanoid glycosides (PhGs) were successfully isolated from Cistanche tubulosa and purified by macroporous resins. This study provides scientific methods for comparing antioxidant activity of BHT, PhGs and VC. A simple and efficient method for large-scale preparative separation of PhGs from Cistanche tubulosa was developed.(1) PhGs were isolated from Cistanche tubulosa by using ethanol. The best parameters of technology were compositely using single-factor test, orthogonal design and Response Surface Methodology. The study with a series of steps including temperature, extraction time, solid to liquid ratio and concentration of ethanol. Experimental results showed that extraction temperature 48 "C,3h of extraction time, 1:12 for the ratio of solid to liquid,83% ethanol, twice extraction. The yield of phenylethanoid glycosides was 3.79%.(2) PhGs were isolated from Cistanche tubulosa by using Ultrasonic-assisted extraction technique. The best parameters of technology were compositely using single-factor test, orthogonal design and Response Surface Methodology. The study with a series of steps including extraction time, Ultrasonic power, solid to liquid ratio, and concentration of ethanol. Experimental results showed that 40min of extraction time,133w of Ultrasonic power,1:18 for the ratio of solid to liquid,81% ethanol, twice extraction. The yield of phenylethanoid glycosides was 4.17%.(3) PhGs were isolated from Cistanche tubulosa by using Microwave assisted extraction method. The best parameters of technology were compositely using single-factor test, orthogonal design and Response Surface Methodology. The study with a series of steps including extraction time, Microwave power, solid to liquid ratio and concentration of Ethanol Experimental results showed that 1.5min of extraction time, 103w of microwave power,1:16 for the ratio of solid to liquid,71% ethanol, twice extraction. The yield of phenylethanoid glycosides was 4.82%.(4) A purification technology for preparative separation of PhGs from Cistanche tubulosa by using macroporous resins was developed. D101 macroporous resin were further screened depending on the adsorption kinetics curves, which showed the best separation efficiency. Experimental results showed that 6h of static adsorption time, the selective adsorption of velocity dynamic adsorption process was 2BV/h, using 80% ethanol as eluent, concentration of sample was 2mg/ml-3mg/ml.(5) This experiment to test the antioxidant activity of BHT, PhGs, VE and VC. the total restoring force:BHT> PhGs>VC; Free radical scavenging activity of them were found to be almost same; Anti-lipid peroxidation ability:PhGs> BHT> VE.
