| With the continuous improvement of living standards,diabetes has become one of the chronic disease killers in the world,in the treatment,although the chemical synthesis of drugs has quick effects,but the side effects still exist;therefore,the health food may has obvious advantages in the treatment of diabetes,which is of interest in future research direction.Literature shows that active ingredients of Wuyi rock tea,Polygonatum and Dendrobium have the function of reducing blood sugar.In this study,Wuyi compound granules were made with Wuyi rock tea as the main raw material,supplemented by tea polysaccharide,huang jing and caulis dendrobii,providing new ideas for prevention and treatment of diabetes.First it is difficult to quantitatively assess the potential hypoglycemic effects of Oolong tea due to its wide variety and abundant active substances..In order to explore a quick and convenient method for evaluating potential hypoglycemic effect of oolong tea,seven kinds of oolong tea samples were tested in China,and mathematical evaluation model of hypoglycemic effects for oolong tea was built using principal component analysis(pca).Activity of alpha glycosidase was determined by measuring generation of pNP with pNPG as substrate.7 oolong tea sample were screened by in vitro screening reaction system for alpha glucosidase inhibitors established with the improvement of traditional pNPG method.The results show that the inhibiting activity of Wuyi rock tea was the highest.In vitro inhibition of alpha glucosidase activity was consistent with the predicted results.The alpha glucosidase inhibitor screening system established in this study has the advantages of simple and rapid,and is suitable for the screening of alpha glucosidase inhibitors in Oolong tea.In vitro hypoglycemic effect of tea polysaccharide,Dendrobium,polygonatum were evaluated with alpha glucosidase activity inhibition rate as the index based on alpha glucosidase inhibitor screening reaction system.And combined with the orthogonal test in vitro synergistic hypoglycemic effects of 3 substances were optimized.The results show that there is a significant synergetic effects with the tea polysaccharide,Dendrobium officinale,and yellow Jingshui extract concentration of 6mg/mL,8mg/mL,and 14mg/mL.Furthermore we blend wuyi rock tea with the optimum formula mentioned above and the compound with a ratio of 3 to 1 for wuyi rock tea and formula has the highest inhibition effects,confirmed by in vitro inhibition experiment of alpha glucosidase activity.Besides,mannitol and gelati were added in the compound using a ratio of 1.5:1.5:1 to make a sweet taste.Finally,rat model of diabetes(rmd)was built by intraperitoneal injection of streptozotocin(STZ)in Wistar rats.And the rmd was treated with wuyi rock tea and wuyi rock tea compound granules of different dosage to measure fasting blood glucose,glucose tolerance,serum glucose(FPG),insulin(INS),triglyceride(TG),free fatty acid(NEFA),superoxide dismutase(SOD),and malondialdehyde(MDA)content after 30d of treatments.Insulin resistance index was calculated according to the results of insulin and blood glucose:pancreatic beta cells were stained with HE and examined by pathology.The results show that the Wuyi Wuyi rock tea and compound granule can improve the tolerance of diabetic rats;decreased serum FPG,TG,NEFA,MDA,increased the content of SOD in diabetic rats;enhance the level of INS and the sensitivity of INS,and decrease INS resistance index.HE staining showed that the number of islets in group of diabetic rats treated by Wuyi compound granule increased gradually,and islet structure was clear with few vacuoles and clear nucleus.Wuyi compound granules on hypoglycemic effect is better than that of Wuyi rock tea.In daily life,taking the Wuyi compound granules can decrease the blood glucose,which is a new health-care products for prevention and treatment of diabetes with a non-toxic side effect. |
