Extraction techniques of Polysaccharide from Inonotus obliquus and purification techniques of alkaloids from Zanthoxylum have been studied in the paper.It has taken various chromatographic technologies to get extracts from the pericarp of Zanthoxylum such as normal and reverse phase silica gel column chromatogram,PTLC,Sephadex LH-20 etc.We have obtained eight compounds.Their structures were identified by various spectral analysis(1H-NMR,13C-NMR,1H-1HCOSY,HSQC,HMBC,NOESY,MS)and three new compounds have been identified.The main results of the study were as follows:1.Studied the extraction techniques of Polysaccharide from Inonotus obliquus extensively and the extraction techniques have been optimized.The heavy metal content of extracts was determined.Aiming at the optimization of extraction technology of Inonotus obliquus polysaccharides on the basis of the one-factor tests,and the method of RSM with 4 factors and 3 levels was adopted.The results indicated that the optimum conditions of Inonotus obliquus polysaccharides extraction as follows:ethanol concentration:31%,extraction temperature:95℃,time of extraction:2.5 h and material to solvent 30:1.Response surface analysis predicted that the production of Crude polysaccharide was 33.6%and the content of polysaccharide was 6.177%.respectively.Crude polysaccharide yield was nearly 2-fold(17%)and the content of polysaccharide increased about 2.9-fold(2.068%)compared to extraction using water.Importantly,application of this method could decrease the concentration of heavy metal significantly.2.Three methods including acidic water method,alcoholic solvent method and acidified ethanol method for extraction of alkaloids from zanthoxylum have been tested.The results showed that the main compositions of the extracts by this three methods had no difference.But the extracting rate was different.The result suggestted acidified ethanol method>alcoholic solvent method>acidic water method.The optimum extraction conditions were ethanol concentration 70%,HC1 0.4%,ratio of solid to liquid 15:1,four times of extractionand in 4 hours.The extraction rate was the highest in the same extraction effect,which is 8.3%.3.Studied the condition of total alkaloids purification of zanthoxylum using macroporous resin.Firstly,alkaloids of zanthoxylum was processed with NKA-9 macroporous resin for absorption and purification,and then,thin layer chromatography and HPLC were used to analyst results.It showed:The optimum elution conditions were washed with 3 BV water and eluted by 50%and 70%ethanol.under adsorption of 4 hours.There were 3 main alkaloids after purification.Those of the peak areas ranged 90%by HPLC.4.The extract of pericaip of Zanthoxylum were isolated using normal and reverse phase silica gel column chromatogram,PTLC,Sephadex LH-20 etc.after the modern spectral analysis(1H-NMR,13C-NMR,1H-1HCOSY,HSQC,HMBC,NOESY,MS),eight amide compounds were identified and three of them are unreported.
