In recent years,traditional Chinese medicine has been widely used in the medical community because of its stable efficacy and low toxicity and side effects.Inonotus obliquus,as a medicinal and food-homogenous fungus in traditional Chinese medicine,can improve hyperglycemia and hyperlipidemia.It has shown an important role in combating metabolic diseases such as obesity and diabetes.Our research group found through literature search that brown fat is an important non-trembling heat-producing organ in mammals,which is rich in mitochondria,can consume energy through direct heat generation,and has a potential role in resisting metabolic diseases.The results of our previous preliminary experiments show that Inonotus obliquus extract can reduce the weight of obese mice induced by high-fat diet.Therefore,this article mainly studies the anti-obesity mechanism of Inonotus obliquus extract and its effect on improving diabetes.The main results are as follows:1.During the period of obesity induced by high-fat diet in mice,the mice were orally administered daily and divided into control groups,125 mg/kg and 250 mg/kg administration groups.Compared with the control group,the mice in the administration group had a significant decrease in body weight and no change in food intake and insulin tolerance was found to be improved by measuring blood glucose.These results proved that Inonotus obliquust extract has anti-obesity effect without changing the food intake.2.In order to determine whether the Inonotus obliquus extract can improve blood lipids,we used kits to test the serum lipids and insulin levels in the serum.The results showed that the Inonotus obliquus extract can improve blood lipids and insulin sensitivity in obese mice.3.In order to further study the mechanism of weight loss of Inonotus obliquus extract,we used qRT-PCR,Western Blot,immunohistochemistry and HE staining to analyze brown fat and related proteins on qualitative and quantitative levels.The results showed that brown fat in mice was obviously activated,and the number of adipocytes became smaller,and white fat appears "brown".UCP1 and heat-related proteins were obviously activated,and fatty liver symptoms had been improved.4.In order to determine whether the weight loss of mice is due to the effect of intestinal flora,we used 16S rDNA V3-V4 regional method to analyze the feces of mice,and confirmed that the administration of Inonotus obliquus extract can significantly improve the composition and structure of the intestinal flora,especially Clostridium papyrosolvens and Enterococcus casseliflavus increased significantly.5.We successfully reproduce the mouse model of type Ⅱ diabetes using the method of high-fat diet combined with STZ.After oral administration of Inonotus obliquus extract,the glucose,glucose tolerance and insulin tolerance of the mice were improved to a certain extent.After testing four serum lipids and insulin content in mice serum,we found that Inonotus obliquus extract can also improve blood lipids and insulin sensitivity in mice.By HE staining,we observed that compared with the model group,the liver and kidney function of the mice in the administration group were improved,and the islet β cell morphology was restored and the number was increased.In summary,this paper confirms that traditional Chinese medicine Inonotus obliquus extract can resist obesity by high-fat diet induced by changing the diversity of intestinal flora and activating brown fat.Besides,Inonotus obliquus extract can also improve the glucose and lipids of type Ⅱ diabetic mice,which provides an important theoretical basis for the development of drugs to resist metabolic diseases.
