Smilax Scobinicaulis C.H.Wright,which belongs to the family of Smilacaceae,is one of traditional Chinese medicine.The clinical use of the roots used as medicine,the study found the rich variety of steroidal saponins in its roots extract.Steroidal saponins have a variety of pharmacological activity and biological effects.For example:promote seed germination,seedling photosynthetic rate and water use efficiency and enhance plant salt tolerance,drought and insect stress.Therefore,how simple,effective from the Smilax extract more steroidal saponins become a hot research.This article is from the goal of total saponin preparation process proceeding to do the following work:1.By using central composite design-response surface methodology to optimize the total saponins in Smilax Scobinicaulis C.H.Wright with UV spectrophotometry.2.The response surface methodology to optimize extraction process of total saponins in the Smilax Scobinicaulis.3.Purification process of tatol saponin from Smilax Scobinicaulis by macro porous adsorption resin.4.Study on purificating the tatol saponin in Smilax Scobinicaulis.The main results are as follows:1.Using central composite design-response surface methodology to optimize the total saponins in Smilax Scobinicaulis C.H.Wright with UV spectrophotometry and to determine the color reagent concentration is 4.8%,reaction time is 10 min,reagent dosage is 2.1 mL to a greater impact factor is the measured absorbance.2.Using central composite design-response surface methodology and single factor to optimize the extraction process of total saponins in the Smilax,obtained the optimum extraction.And to determine the 64%of the ethanol concentration,extraction time of 92min,solid to liquid ratio of 1:8 is the best extraction conditions。3.Study on purification process of tatol saponin from Smilax Scobinicaulis by macro porous adsorption resin.In this paper,the adsorption and elution of the three kinds of macro porous resin is studied.Eluted saponin content as an index of the best resin adsorption and elution conditions were selected.Selected of SP-825 resin for the purification of the best resin and drawing the best adsorption is 2.50mg/mL and elution parameters by the sample volume is 2 BV/h,flow rate concentration of eluting solvent is 50%ethanol and elution dosage is 5BV.4.The root powder was extracted using the optimized extraction process.The concrete was extracted respectively with petroleum ether,ethyl acetate and n-butyl alcohol.N-butanol fraction preliminary purificated by the macroporous resin,then 95%ethanol fraction separated by silicagel column,some components repeatedly recrystallized.The method is simple,total saponins have high purity.This method offer a valid way to production total saponins in Smilax Scobinicaulis.
