China is rich in edible mushroom resources,and the exploitation of wild edible mushrooms has important value to enrich the edible mushroom resources in China.In this study,a wild strain of Hohenbuehelia geogenia collected at Tibet Agricultural and Animal Husbandry University was used as the test material,and the wild fungus was studied through isolation and identification,biological characterization,enzyme activity determination,inhibition test study and domestication cultivation test.To lay the foundation for the subsequent study of this wild fungus.The main research results are as follows:(1)Pure culture of strain HGHZ02 was obtained by tissue isolation,and although ITS sequencing results showed that strain HGHZ02 was100%similar to Hohenbuehelia petalodes and obtained 100%support,morphological characteristics showed that strain HGHZ02 and H.petalodes had However,the morphological characteristics showed that there were significant differences between strain HGHZ02 and H.petalodes,and its morphological characteristics were more consistent with those of H.geogenia.Through the combination of morphological identification and molecular biological identification,strain HGHZ02 was finally identified as H.geogenia.(2)Through the plate culture method,the strain HGHZ02 was screened for the parent culture medium,and four different parent culture mediums were set up for the test.Taking into account the mycelial growth rate,growth potential and ANOVA of the data,the most suitable parent culture medium for the mycelial growth of strain HGHZ02 was PDA.(3)The biological characteristics of strain HGHZ02 were studied by plate culture method,the optimum carbon source for mycelial growth of strain HGHZ02 was maltose,the optimum nitrogen source was malt extractpowder,the optimum temperature was 25℃,the optimum p H 7.0,the optimum inorganic salt was K2HPO4,and the optimum growth factor was vitamin C.The orthogonal test results showed that the optimum combination of factors for strain HGHZ02 was maltose 25.0 g/L,malt extract 3.0 g/L,p H 7.0 and temperature 26℃.(4)The enzyme-producing activities of strain HGHZ02 were determined by liquid culture method,and the results showed that the highest enzyme activities of carboxymethyl cellulase,laccase,amylase and xylanase were 69.62 U/m,21.77 U/m L,44.70 U/m L and 207.13U/m L,respectively,with the highest xylanase activity.(5)The inhibition of strain HGHZ02 was determined by the plate confrontation method,a total of 16 pathogenic fungi were selected,and the results of the initial and re-screening tests showed that strain HGHZ02 showed weak inhibition effect on Bipolaris maydis and Rhizoctonia solani,with inhibition rates of 17.38%and 14.57%.(6)The domesticated cultivation of strain HGHZ02 was explored,and 11 cultivars were designed for the domestication of strain HGHZ02.The results showed:strain HGHZ02 grew strongly in optimized formulations 8,9 and 10,with mycelial growth rates of 3.11 mm/d,2.90mm/d and 2.81 mm/d,respectively,but no protoplasts appeared during the cultivation process,and further studies are needed.In conclusion,H.geogenia is a high quality edible fungal germplasm resource with potential for exploitation,and at the same time,this study lays the foundation for the exploitation of H.geogenia strain.
