| With the development of intensive and large-scale farming,the prevalence of metabolic diseases is increasing,which seriously affects the economic benefits of animal husbandry.The liver is an essential organ for regulating lipid metabolism in livestock and poultry.Disturbance of lipid metabolism in the liver can cause a large amount of fat deposition and induce fatty liver syndrome.Solanesol(SNL)is a terpenoid compound with strong antioxidant capacity and can reduce lipid peroxidation.The purpose of this thesis is to study the effects of solanesol on liver lipid metabolism and its possible mechanism.Firstly,an animal model of fatty liver syndrome was established by feeding yellowfeathered broilers with a high-fat diet,and the regulation effect of solanesol on liver lipid metabolism was analyzed.Secondly,oleic acid/palmitic acid(OA/PA)was used to induce fat deposition in human hepatoma cells(Hep G2),to study the effect of solanesol on intracellular lipid metabolism-related indicators,and to explore its possible molecular targets.The main results are as follows:At the in vivo level,solanesol ameliorates liver lipid deposition in broilers induced by high-fat diets.Solanesol reduces liver triglyceride(TG)and serum TG,total cholesterol(TC)and aspartate aminotransferase(AST)levels and relieve overweight and regulate muscle quality of broilers induced by high-fat diet.At the cellular level,solanesol ameliorated OA/PA-induced TG accumulation in hepatocytes and reduced oxidative stress through β-oxidation.Solanesol can reduce the levels of TG and TC in OA/PA-induced hepatocytes,up-regulate the m RNA expression levels of β-oxidation-related genes,and increase the activity of short-chain acyl-Co A dehydrogenase(SCAD).Solanesol also enhanced the activity of superoxide dismutase(SOD)in liver cells,increased the content of glutathione(GSH)and decreased the level of reactive oxygen species(ROS).In addition,the results of network pharmacology analysis revealed that the key targets of solanesol regulating lipid metabolism may be Pparα,Akt1 and Pparγ.This study reveals the prevention and treatment effect of solanesol on chicken fatty liver syndrome,and provides a new direction for the treatment of livestock and poultry nutrition and metabolism related diseases.At the same time,the research results provide experimental evidence for the development of solanesol as a new feed additive. |
