| Apodemus peninsulae and Apodemus agrarius are common rodent species in northeastern China and are the focus of rodent control.Related studies have shown that the intestinal flora of the two rodent species are somewhat different from each other,but there are not enough studies on whether and what effects habitat factors have on the intestinal flora between the two rodent species.In this paper,we captured Apodemus peninsulae in mixed coniferous and broadleaf forest habitats and Apodemus agrarius in scrub and farmland habitats,and performed species annotation and taxonomic analysis,Alpha and Beta diversity analysis,between-group difference significance analysis,correlation and association analysis,Bugbase phenotype prediction,and intestinal flora metabolic function prediction by using the more mature second-generation sequencing technology.We analyzed whether habitat factors had an impact on the intestinal flora of the two species and whether the differences were significant,whether the structural composition of the intestinal flora of the same species differed between habitats,and what aspects of the flora function of the intestinal flora of the same species differed between habitats.The results of the study are as follows.The results of the study were as follows:(1)The first two dominant phyla of DLH(Apodemus peninsulae in mixed forests)and DLK(Apodemus agrarius in broad-leaved forests)were both Phylum Firmicutes and Phylum Bacteroidota,and the first three dominant phyla of HXG(Apodemus agrarius in scrub)and HXN(Apodemus agrarius in agricultural fields)were consistent,being Phylum Firmicutes,Phylum Bacteroidota,and Phylum Proteobacteria,respectively.(2)Beta diversity analysis showed significant groupings among the four groups and moderate differences among the groups.(3)Using LEf Se analysis,it was found that HXG,DLK and DLH all had at least one genus and species significantly different from the other 3 groups of samples(P<0.05).(4)When Bugbase phenotype prediction was performed,when samples were compared two by two,among the nine phenotypes tested,three pairs of samples,DLH and DLK,DLK and HXG,and DLK and HXN,showed significant differences(P<0.05)for he two phenotypes of Facultatively Anaerobic and Forms Biofilms,and only DLK and HXG showed significant differences(P<0.05)for the three phenotypes of Gram Negative,Gram Positive and Potentially Pathogenic,while the remaining four phenotypes did not show significant differences among the four groups of samples.(5)The prediction of metabolic functions of intestinal flora based on KEGG database showed that the annotation of the first functional layer showed that there were significant differences(P<0.05)in at least one function of Cellular Processes between Apodemus peninsulae and Apodemus agrarius and between Apodemus agrarius in different habitats;the annotation of the second functional layer showed that there were significant differences(P<0.05)in metabolic functions between Apodemus peninsulae and Apodemus agrarius,and also due to the effect of habitat The metabolic function inconsistency between species was significant.In summary,although the dominant phylum in all four groups of samples was dominated by Phylum Firmicutes and Phylum Bacteroidota,there were disparities in species and order.Predictions of phenotypic and metabolic functions showed inconsistent predictions for the same gypsy in different habitats,and habitat factors could affect the structural composition of the gut flora in both gypsy species. |
