| Aegilops geniculata Roth(Aegilops geniculata Roth,2n=4x=28,UgUgMgMg)represents a source of potentially useful genetic variation of relevance to bread wheat improvement,its high molecular weight glutenin subunits on 1Mg chromosome are valuable in wheat breeding.In this study,screening population and markers were developed to obtain to wheat translocation line carrying 1Mg glutenin gene by centric breakage and fusion.The main results are as follows:1.The 1Mg and 1A centric breakage and fusion translocation lines screening population[CSN1AT1B/(add L-1/CSph1b)F2]was constructed by hybridization and molecular marker selection.2.Referring to the homologous gene sequence of PLUG marker,6 KASP markers on short arm of 1Mg(0.44M on 1A)and 2 KASP markers on long arm of 1Mg(381.4M on 1A)were developed.1 KASP markers of 1Mx was also developed.3.Molecular identification showed that four plants(B1,B9,A1 and A22)carried only 1MgL and 1AS chromosomes,and their seeds of transmissibility of 1My HMW-GS were 40%-48%respectively,1AS.1MgL translocation may have occurred in these plants.A1 has homozygous ph1b and can be used to screen small fragment translocation of 1MgL in next generation.In addition,there are another 2 plants(A15and A18)have homozygous ph1b,their seeds may have small fragment translocation of 1Mg L.4.T1AS.1MgL translocation lines A1-65 was identified by FISH karyotype. |
