| High-molecular-weight glutenin subunits(HMW-GSs)are one of the most important proteins on the processing quality of wheat flour.In our previous work,we identified and cloned two novel HMW-GS genes of the x-type and the y-type from Aegilops sharonensis(Ae.Sharinensis),and have successfully introgressed them into common wheat.We sequentially conducted backcrosses,expected to get the lines containing alien HMW-GSs from Ae.sharonensis with good agronomy traits and genetic stability.The 24 derivatives with homozygosis types of HMW-GS were screened out from the wheat-Aegilops sharonensis derived lines by using SDS-PAGE and cytological analysis.The results are listed as following:1.The investigations of agronomic traits revealed that the derivatives showed a stable tendency.There was a significant increase in setting rate for six combinations,such as the combination of the cross between Z636 and R7,and then backcross with L3.The setting rate increased from 14.71%to 41%,the seeds number rose from 10 of BC1 to 121910 of BC6.On the other hand,eight combinations has become extinct because of barrenness.2.HMW-GS compositions were checked by sodium dodecyl-sulfate polyacrylamide gel electrophoresis(SDS-PAGE),and 24 lines containing novel HMW-GSs from Ae.sharonensis with genetic stability has been characterized.They all originated from the same combination of the cross between Z636 and R7,and then backcross with L3.The compositions of the HMW-GS are as follows:(1)containing Aegilops sharonensis,the tetraploid wheat of all subunits and the D-subunit of liangmai 3;(2)all subunits of Aegilops sharonensis and Liangmai3,have been lost tetraploid’s;(3)all subunits containing the Aegilops sharonensis’s and B subunit of liangmai3;(4)all subunits containing the Aegilops sharonensis’s and the D subunit of liangmai 3.Sixteen of 24 lines have relatively higher setting rate,5 strains was over 90%and 6 strains was over 70%.3.The chromosome compositions of root-tip cells of the 24 lines were observed,and the cytological analysis showed that chromosome numbers ranging from 41 to 44.Nine of the lines have 42 chromosomes,three have 40 chromosomes,two have 41 chromosomes,and three have 44 chromosomes. |
