Tomato is an important cash crop in China.Tomato chlorosis is a virus disease of tomato caused by Tomato chlorosis virus(To CV)that is widely distributed worldwide.After its first occurrence in Taiwan,it rapidly spread throughout the country’s provinces,causing serious impacts on tomato production.To CV is transmitted semi-persistently mainly by vector insects such as whiteflies,and most of the existing control measures rely on the control of insect vectors to control the spread of the virus,and there are no To CV-resistant varieties on the market.In recent years,photosynthetic bacteria have become a popular biological agent due to their environmental friendliness and rich nutrient content,and some reports have shown that photosynthetic bacteria are effective against tomato virus disease.In this study,we tested the efficacy of Rhodopseudomonas palustris PSB-06 on To CV in the field for three consecutive years,the changes in the content of To CV in tomato plants indoors by q RT-PCR,the effects of PSB-06 on the survival and behavioral activities of whiteflies were investigated by selective tests and the proteomic analysis of PSB-06-treated tomato plants was performed.The aim of this study was to determine the inhibitory effect of Rhodopseudomonas palustris PSB-06 on To CV.The main results are as follows:1.PSB-06 treatment could reduce the occurrence of To CV,and the incidence of plant disease in all three years was lower than 50%,which was significantly lower than that of water and 5% amino-oligosaccharide treatment;the disease index was lower than20%,which was significantly lower than that of water and 5% amino-oligosaccharide treatment;while the control effect was higher than 70%,which was significantly higher than that of 5% amino-oligosaccharide treatment.2.The number of MED on upper tomato plants infected with To CV after 24,48 and72 h of spraying with Rhodopseudomonas palustris PSB-06 fungicide was counted,and the results showed that MED preferred tomatoes treated with water during the three observation periods;meanwhile,the survival rate and fecundity of whiteflies on PSB-06-treated plants were lower than those of the control,with the most significant difference on the third day,indicates that PSB-06 reduced the preference,survival and fecundity of whiteflies.3.The graft inoculation method was effective in transmitting the virus,and the amount of virus on tomatoes after grafting was significantly reduced by PSB-06 fungicide treatment,indicating that PSB-06 treatment could reduce the virus accumulation in tomato plants.4.The results of proteomic analysis showed that PSB-06 treatment induced upregulation of proteins related to both plant metabolism and defense.This indicates that Rhodopseudomonas palustris PSB-06 can enhance plant immunity and defense,thus effectively controlling the occurrence and spread of To CV.
