important part of the gastrointestinal tract function of the organism.Intestinal bacteria and fungi interact with each other to maintain intestinal homeostasis.Intestinal flora plays an important role in physiological regulation,metabolism and immunity,and is closely related to the growth,development and health of the organism.Although many studies have shown that TCM polysaccharides have certain regulatory effects on the intestinal flora of cultured fish,the mechanism of TCM polysaccharides in regulating Luciobarbus capito intestinal flora remains unclear.In view of this,combined with the previous research experience of the research group,this study intends to select Yam polysaccharide(YP)and Lycium barbarum polysaccharide(LBP)respectively to analyze the composition,structure and dynamic changes of intestinal flora after Luciobarbus capito treatment.At the same time,the growth performance and biochemical indexes of Luciobarbus capito before and after treatment of Yam polysaccharide and Lycium barbarum polysaccharide were combined to provide a theoretical basis for the healthy cultivation of Luciobarbus capito.The treatment of Yam polysaccharide and Lycium barbarum polysaccharide can obviously improve the structure of Luciobarbus capito intestinal flora,promote the growth of Luciobarbus capito and significantly improve the digestive performance and immunity.The relevant results of this study are not only helpful to the development and utilization of saline-alkali water and saline-alkali soil resources in China,but also appropriate consumption of Luciobarbus capito is helpful to human health and prevent the occurrence of chronic diseases such as cardiovascular disease and liver disease.It provides experimental basis and theoretical guidance for the regulation of Luciobarbus capito intestinal flora by TCM polysaccharides,the improvement of immunity and the development and application of new feed additives.Better complete the ecological aquaculture goal of "treating alkali by fishing".Relevant results of this study are described as follows:1.Effect of Yam polysaccharide on intestinal flora of Luciobarbus capitoThis study investigated the effects of different doses of Yam polysaccharide(0.10 g/L,1.00 g/L and 10.00 g/L YP)on intestinal flora,digestive capacity,immunity and antioxidant properties of Luciobarbus capito.It was found that 0.10 g/L YP treatment could significantly reduce the relative abundance and diversity of intestinal flora in Luciobarbus capito(P>0.05),and the relative abundance of Proteobacteria was slightly higher than that in control group(P>0.05).The relative abundance of Ascomycota was significantly lower than that of the control group(P<0.05),while the relative abundance of Basidiomycoat was significantly increased(P<0.05).The results of enzyme activity analysis showed that the activities of digestive enzyme,antioxidant enzyme and immune enzyme of Luciobarbus capito were significantly increased(P<0.05).These results indicated that 0.10 g/L yam polysaccharide could significantly change the intestinal flora structure of Luciobarbus capito and enhance the digestive ability,immunity and antioxidant properties of Luciobarbus capito.2.The effects of Lycium barbarum polysaccharide on intestinal flora of Luciobarbus capitoThis study further explored the effects of different doses of Lycium barbarum polysaccharides(0.10 g/L,0.50 g/L and 1.00 g/LLBP)on the intestinal flora,digestive capacity,immunity and antioxidant properties of Luciobarbus capito,and found that the intestinal microbial composition of Luciobarbus capito changed significantly before and after LBP treatment.Their diversity and abundance were generally decreased(P>0.05).After 0.50 g/L LBP treatment,the relative abundance of Proteobacteria Luciobarbus capito reached the peak,while the relative abundance of A eromonas was slightly lower than that of control group(P>0.05),and the relative abundance of Acremonium was slightly higher than that of control group(P>0.05).The results of enzyme activity analysis showed that 0.50 g/L LBP treatment significantly increased the activities of digestive enzymes,antioxidant enzymes and immune enzymes of Luciobarbus capito(P<0.05).These results indicated that Lycium barbarum polysaccharide could significantly improve the intestinal flora structure of Luciobarbus capito and enhance the digestive performance,immunity and antioxidant properties of the body,and the optimal concentration of Lycium barbarum polysaccharide was 0.50 g/L.3.Effects of Yam polysaccharide and Lycium barbarum polysaccharide on the growth,Serum Biochemical indexes and Immune genes of Luciobarbus capitoThis study further explored the effects of YP and LBP on growth performance,serum biochemical indices and immune genes of Luciobarbus capito.YP group was divided into 0(control),0.10%,0.20%,0.40%and 0.80%YP groups.The LBP group was divided into 0(control),0.10%,0.20%and 0.40%LBP groups.Each group had 3 replicates.The experiment lasted for eight weeks.The results showed that the weight,weight gain rate and specific growth rate of Luciobarbus capito before and after 0.40%YP treatment were significantly higher than those of control group(P<0.05),and the feed conversion ratio was significantly lower than that of control group(P<0.05).The urea nitrogen content of 0.40%YP and 0.80%YP treatment groups was significantly lower than that of the other three groups(P<0.05);The SOD gene expression level in 0.40%YP treatment group was significantly higher than that in the other four groups(P<0.05).In contrast,the mass and weight gain rate of 0.10%LBP treatment group was significantly higher than that of the other three groups(P<0.05),and the viscerosomatic ratio of control group was significantly higher than that of 0.10%and 0.40%LBP treatment groups(P<0.05).The triglyceride in 0.10%LBP group was significantly lower than that in control group and 0.40%LBP group(P<0.05).The urea nitrogen level in 0.40%LBP group was significantly lower than that in control group(P<0.05).AKP gene expression in control group was significantly higher than that in other three groups(P<0.05).These results indicate that 0.40%polysaccharide can significantly improve the growth performance and gene expression of Luciobarbus capito.Adding 0.10%Lycium polysaccharide in the diet can significantly improve the growth performance of Luciobarbus capito and has the effect of lipid lowering. | | Keywords/Search Tags: | Yam
