| Immunosuppression refers to a temporary or permanent state of immune dysfunction caused by infection,stress,abuse of antibiotics and chemicals and other factors.Animals in this state may increase the incidence of secondary infection and immune deficiency,causing great losses to the poultry industry.At present,modern pharmacological studies have confirmed that Lycium barbarum polysaccharide(LBP)has antioxidant,anti-inflammatory,neuroprotective,hypoglycemic and reducing blood sugar and lipid levels and immune regulation functions,but whether it has an antagonistic effect on immune suppression of chicks needs to be further verified.Therefore,the antagonistic effect and mechanism of LBP on immunosuppression of chicks were studied.1.A total of 200 7-day-old Hyline brown laying hens were randomly divided into blank group(NC),cyclophospamide group(CY),high dose lycium barbarum polysaccharide group(LbGpH),medium dose lycium barbarum polysaccharide group(LbGpM)and low dose lycium barbarum polysaccharide group(LbGpL),with 40 hens in each group.After the successful modeling,10mg/kg·d LBP of LbGpH group,5mg/kg·d LBP of LbGpM group and 2.5 mg/kg·d LBP of LbGpL group were given water for 7 consecutive days.On days 7,14,21 and 28 after administration,5 chickens in each group were randomly selected to measure body weight,spleen index,peripheral blood lymphocyte transformation,and spleen histological observation.The results showed that LBP could significantly improve the body weight,spleen index and peripheral lymphocyte conversion rate of immunosuppressive chicks,and improve the pathological injury of the spleen.2.120 7-day-old Hyline brown laying hens were randomly divided into Negative control group(NC),Cyclophosphamide group(CY)and Lycium barbarum polysaccharide group(CYLbGp).The NC group was intramuscularly injected with normal saline,and the other two groups were intramuscularly injected with 80 mg/kg·d CTX for 3 consecutive days to establish disease model.Then 5 mg/kg·d LBP was added into the drinking water of LBP group for 30 consecutive days.Spleens of 6 chicks in each group were collected for transcriptome analysis.The results showed that LBP intervention after a total of 178 genes significant difference correction,significantly enriched in Mitophagy-animal,Chemokine signaling pathway,C-type lectin receptor signaling pathway,B cell receptor signaling pathway,Platelet activation,Natural killer cell-mediated and cytotoxicity,Thl and Th2 cell differentiation and other pathways.3.In this study,UHPLC-triple-TOF-MS metabolomics technique was used to study the effects of LBP on metabolites in the spleen of immunosuppressive chicks.A total of 20 different callback metabolites were identified,mainly involving vitamin metabolism,lipid metabolism,nucleic acid metabolism and amino acid metabolism,and the analysis in transcriptome and metabolome showed that glycerol phospholipid metabolic pathway was the most important metabolic pathway.Conclusion:LBP has a good antagonistic effect on immunosuppression of chicks,and the 5 mg/kg·d lycium barbarum polysaccharide supplemental group has the best therapeutic effect.LBP can improve immune function by regulating Mitophagy-animal,Chemokine signaling pathway,C-type lectin receptor signaling pathway,B cell receptor signaling pathway,Platelet activation,Natural killer cell-mediated and cytotoxicity,Thl and Th2 cell differentiation and other pathways.The antagonistic mechanism of LBP against immune suppression in chicks may be closely related to its correction of vitamin metabolism,lipid metabolism,nucleic acid metabolism and amino acid metabolism disorders. |
