| In order to explore the potential molecular mechanisms related to the meat quality and fat deposition of Debao and Changbai pigs.In this project,the subcutaneous visceral fat of6-month-old Debao pigs and Changbai pigs was selected as the research object.Transcriptome sequencing technology was used to explore different adipose tissues and the data of sequencing data was used to deeply explore the genes with differential expression at the transcription level.Subsequently,relevant GO function annotations and KEGG enrichment analysis were carried out on these data,and finally the accuracy of the transcriptome data was verified using fluorescence quantitative experimental methods.The results obtained in this project are as follows:1.The ratios of clean reads of Debao pig and Landrace pig to the reference genome are:93.96,93.82,94.95,95.36;2.Through saturation analysis,it was found that when the reads in 4 samples exceeded80 M,the curve tended to be flat,indicating that it reached saturation,and the number of clean reads obtained through screening exceeded 100 M,indicating that 4 samples met the subsequent requirements.3.In the principal component analysis of 4 samples of two pig breeds,the Pearson coefficient between the 4 samples was found to be above 95%,indicating that the 4 samples had good repeatability.4.Set the conditions for screening differential genes: q value <0.001,|log2FC|>=3,obtain genes with significant differences.The differential genes between the four groups of samples are: DB-VF and CB-VF.There are 932 differentially expressed genes in the two groups of samples,of which 235 genes are up-regulated and 737 genes are down-regulated;DB-SF and There are 1088 differentially expressed genes in the CB-SF two groups of samples,including772 genes that are up-regulated and 316 genes that are down-regulated;432 differentially expressed genes in the DB-VF and CB-SF samples Among them,there are 217 genes that are up-regulated and 215 genes that are down-regulated;there are 331 differentially expressed genes in DB-SF and CB-VF samples,of which 103 are up-regulated and down-regulated genes There are 228.5.Relevant GO function annotations and KEGG enrichment analysis of these differential genes to further obtain the metabolic pathways involved in the core genes and the functions they perform.It is found that these core genes mainly perform metabolic functions,biological regulatory functions,catalytic functions,etc.Etc.;through in-depth analysis of the metabolic pathways involved,it is found that it is mainly related to metabolic related pathways,such as:sugar metabolism,pyruvate metabolism,carbon metabolism pathways,etc.Through analysis and in-depth mining of related pathways,some core genes were found,including: CERS1,AKR1C4,GADL1,MDH2,ACAT2,DGKI,COQ7,PKLR,COX5 B,SMPD3,LARGE2 and so on.6.Among the genes with significant differences,8 genes were randomly selected for fluorescence quantitative verification.It was found that there was no significant difference between the expression results of these 8 genes and the transcriptome data,indicating that the transcriptome data was very reliable and correct.In a short,this experiment obtained transcriptome sequencing of 4 adipose tissue samples from two pig breeds to obtain the expression of many genes related to fat metabolism.Through in-depth mining of transcriptome data,the core genes of different pig breeds and different adipose tissues and the signal pathways involved are found,which provides more rigorous scientific data for the molecular breeding of Debao pigs in the future. |
