Objective:To construct the rat model of acute soft tissue injury, to observe the morphological changes of soft tissue injury in rat; and to observe the effect of different treatment methods on rat acute soft tissue injury in subacute stage, and explore the possible mechanisms, in order to provide according treatment to clinical medication.Methods:A total of60postnatal Wistar rats were randomly divided into four groups, Control group(CG), Icing group(IG), Hyperthermia group(HG), Heat and cold alternating group(HCG). Using the self-made device strikes the inner thigh soft tissue in each group, in order to construct the rat model.Feeding them in nature light environment. After48hours, the rats in IG were iced with4℃biological ice pack, the rats in HG were heated with51℃biological hot pack, and the rats in HCG were dealed with51℃biological hot pack and4℃biological ice pack interval of5minutes alternately, and2hours total. The rats in CG didn’t do any disposal.72hours later, all of the rats were executed, and were to selected injury tissue, the injury tissue were made into homogenate, in order to determine the superoxide dismutase(SOD) and malondialdehyde(MDA) in the injury tissue. And the remainder muscles and subcutaneous were made into paraffin secetion, to observe the muscle fiber changes and inflammatory cell infiltration, and the expression of transforming growth factor-β1(TGF-β1) as well was detected by immunohistochemistry.Results:Compared with the CG, the histological grading were significantly reduced in HG and HCG (P<0.05); while the expression of TGF-β1and SOD were increased, and the expression of MDA was reduced (P<0.05). Compared with the HG, HCG showed higher expression of TGF-β1and SOD, and lower expression of MDA (P<0.05).Conclusion:①Hyperthermia, alternating hot for subacute phase of soft tissue injury in rats can get treatment effectiveness, and the alternating hot and cold treatment is better than Hyperthermia.②which is likely to reach the goal through raising by raising TGF-β1and SOD expression, and lowering MDA expression.
