Effects Of Jieqi Capsules On IL-1β, SOD Content And Caspase-3 Expression In Rats With Acute Soft Tissue Injury

Objective: By observing the rat model of acute soft tissue injury,at different time points,the skin temperature,the index of injury symptom,the content of IL-1β and SOD in serum,the expression of Caspase-3 activity in injured muscle tissue were recorded,to investigate the mechanism and mechanism of action of Jieqi capsules in the treatment of acute soft tissue injury,and to provide basic experimental reference for clinical application.Methods: The study involved 72 adult male Sprague-Dawley rats at 8 weeks of age.The rats were randomly divided into three major groups(24 rats per major group)according to the Rand function in Excel by numbering,namely,group A(Jieqi),group B(Qili),and group C(Saline).The rats were moulded using the local heavy blow method and the skin temperature difference and injury syndrome index were recorded at 1,3,7 and 10 days after moulding;Measurement of SOD and IL-1β in serum by Elisa method;The local injury tissues were taken,caspase-3 activity was detected by spectrophotometric method and caspase-3 expression was detected by immunohistochemistry.The data were analyzed using spss22.0 statistical software,and the influence of Jieqi capsules on the change of indicators was observed by comparing between and within groups,and a significance level of P<0.05 was set as statistically significant.Results:(1)Skin temperature difference: The difference in skin temperature between the major groups is inversely proportional to the increase in the number of days.1 day after moulding,the data show that there is basic agreement among the major groups(P>0.05).After 3,7 and 10 days of moulding,at different points in time,the saline group was higher than the Jieqi and Qili groups,there was a significant difference(P<0.05).(2)Injury Symptom Index(ISI):The three major sets of indices are inversely related to time.After 1 day of modelling,the data showed that there was no significant difference among the groups(P>0.05).Three days after modeling,the saline group index was significantly higher than that of the Jieqi group,and there was a significant difference(P<0.05).After 7 and 10 days of modeling,there were significant differences between the salt water group and the Jieqi group and the Qili group respectively(P<0.05).(3)SOD content: The SOD content within the serum of the major groups was directly proportional to the prolongation of the days.At different time points,the SOD levels in the Jieqi and Qili groups were significantly better than those in the saline group,with a large difference(P<0.05).(4)IL-1β levels and caspase-3 activity: serum IL-1β levels and caspase-3 activity in the major groups rose to a peak on day 3 and then showed a decreasing trend,with timeand drug-dependent characteristics.One day after modeling,there was little difference between the major groups(P>0.05).After modeling 3,7,and 10,there were significant differences between the salt water group and the Jieqi group and the Qili group respectively(P<0.05).(5)Caspase-3 immunohistochemistry: Under the intervention of Jieqi Capsule,the expression of Caspase-3 can be regulated to greatly express in injured muscle cells and inflammatory cells,reducing its expression in healthy muscle fibres as well as cells with phagocytic capacity,accelerating the clearance of necrotic tissues and achieving the promotion of recovery of damaged tissues.Conclusion:(1)Jieqi capsule can improve the skin temperature and swelling of the injured limb after acute soft tissue injury in rats,and accelerate the recovery of the injured body.(2)the mechanism of Jieqi capsule in the treatment of acute injury may be related to regulating the activity of Caspase-3 in injured tissue,promoting the expression of Caspase-3 in injured muscle fibers,reducing the apoptosis of phagocytes and accelerating the apoptosis of injured cells.(3)the therapeutic mechanism of Jieqi capsule may be related to reducing the content of IL-1β and increasing the content of SOD.The ultimate goal is to accelerate the healing of injured tissues,which has a high clinical application value for acute soft tissue injury repair.
