Effects Of Zanthoxylum Bungeanum Xinshang Oil On PGE2 Content And COX-2, MPGES-1 MRNA Gene Expression In Rats With Acute Soft Tissue Injury

Objective: Through the establishment of acute soft tissue injury model in rats,the skin temperature of the affected area,soft tissue injury symptomology,local muscle tissue morphological changes,PGE2 content and COX-2,mPGES-1 mRNA gene expression values were observed and detected before and after the medication,and the new injury of Zanthoxylum bungeanum was preliminarily discussed.The effect of oil on acute soft tissue injury in rats and its possible mechanism of action.Methods: Sixty SD rats(including 6 spares),8 weeks old,male,adaptively fed for 1 week were divided into 3 groups according to the random number table method,each with 18 rats;SD rats were made by "quantitative gravity strike method" After the acute soft tissue injury model in rats,group H(Zanthoxylum bungeanum oil group): apply Zanthoxylum bungeanum oil for external application;group F(excipient group): apply excipient for external application;group K(blank control group): No model and no drugs were used;each group was randomly divided into the 1st day group(n=6),the 4th day group(n=6),and the 7th day group(n=6)according to different time points;observation and The rats’ injury recovery and histopathological changes,PGE2 content in muscle tissue and COX-2 and mPGES-1 mRNA gene expression values were detected on the first 1,4,and 7 days after the medication;SPSS 20.0 software was used to analyze the data.Results:(1)Skin temperature comparison results.Intra-group comparison: After application,the bilateral skin temperature difference of rats decreased with time.The F and H groups had significant differences on the 1,4,and 7 days of each group(P<0.05).);comparison between groups: on days 1,4,and 7 the skin temperature of the F and H groups were higher than that of the K group,and there was no significant difference in the skin temperature difference between the F and H groups on the 1st day(P>0.05).Significant difference in comparison between 4 and 7 days(P<0.05);(2)Comparison of histology and slice results within groups: There was no significant difference in the comparison between the first and fourth days in group F(P>0.05);the comparison between the first and seventh days was Significant difference in comparison between the 4th and 7th day(P<0.05);Significant difference in the relative comparison of the H group on day 1,4,and 7(P<0.05);Comparison between groups: No comparison between H and F groups on day 1 Significant difference(P>0.05);on the 4th and 7th day,the H and F groups were significantly different(P<0.05);the K group was significantly different from F and H on the 1,4,and 7 days(P<0.05)both Under light microscope observation,the muscle tissues of F and H groups had different degrees of muscle fiber tissue degeneration and necrosis,inflammatory cell infiltration,etc.,with the extension of the application time,the H group was less swollen than the F group.Faster muscle fiber repair;(3)Results of changes in PGE2 content.Comparison between groups: F and H groups on day 1,4,and 7 have significant differences in pairwise comparisons(all P<0.05);comparison between groups: group K on day 1 Compared with the F and H groups,the difference was significant(P<0.05),and there was no significant difference between the F and H groups(P>0.05);on the 4th day,the K group was significantly different from the F and H groups(P<0.05),F and H groups >0.05);(4)Comparison of COX-2 mRNA gene expression results within groups: F and H groups were significantly different on day 1,4,and 7(all P<0.05);comparison between groups: group K and group K on day 1 There was a significant difference between F and H(P<0.05),and there was no significant difference between F and H groups(P>0.05);on day 4,there was a significant difference between K and F groups(P<0.05),and K and H groups were similar.Compared with no significant difference(P>0.05),there is a significant difference between F and H groups(P<0.05);on the 7th day,there is a significant difference between K and F groups(P<0.05),and there is no difference between K and H groups.Significant difference(P>0.05),the difference between F and H groups was significant(P<0.05);(5)Results of mPGES-1 mRNA gene expression value comparison within groups: F and H groups on the first 1,4,and 7 days The difference between the two groups was significant(P<0.05);the comparison between the groups: on the first day,the K group was significantly different from the F and H groups(P<0.05);there was no significant difference between the F and H groups(P >0.05);On the 4th day,the K group was significantly different from the F and H groups(P<0.05),and the F and H groups were significantly different(P<0.05);the K and F groups were significantly different on the 7th day(P<0.05),K,There was no significant difference compared with H(P>0.05),and the difference between F and H groups was significant(P<0.05).Conclusion: 1.External application of Zanthoxylum bungeanum oil to treat acute soft tissue injury in rats can improve the injury symptoms and tissue morphology of rats,and accelerate the repair of damaged tissues.2.External application of Zanthoxylum bungeanum oil to treat acute soft tissue injury in rats can reduce the content of PGE2 in damaged tissues and inhibit the expression of COX-2 and mPGES-1 mRNA in damaged tissues.3.The anti-inflammatory mechanism of Zanthoxylum bungeanum oil in the treatment of acute soft tissue injury in rats may be related to the COX-2/mPGES-1/PGE2 pathway.By inhibiting the expression of COX-2 and mPGES-1 mRNA in local tissues,it reduces PGE2 Produce and play an anti-inflammatory effect.
