Objective:To observe the effects of pomegranate peel polyphenol cream of different concentrations on the size and morphology of sebaceous glands of golden hamster and the expression of PPARγ/ABCA1 signaling pathway in each group,and to explore the mechanism of pomegranate peel polyphenol cream on the regulation of sebum metabolism in acne.Methods:36 SPF golden hamsters were divided into blank group(model group),PPARγ agonist(rosiglitazone)group,positive drug(retinoic acid cream)group and pomegranate peel polyphenol cream low-concentration,medium-concentration and high-concentration groups,with 6 in each group.The blank group did not intervene.Rosiglitazone group was given 4 mg/(kg·d)intragastric gavage,once a day;The positive control group and the pomegranate rind polyphenol cream group were given 0.025%retinoid acid cream and 1g pomegranate rind polyphenol cream(self-made low,medium and high concentration)topically applied to the sebaceous gland spots on the back of golden hamster,twice A day.Intervention intervention before and 4 weeks after measured the golden mouse back sebaceous glands spot area,in order to the longitudinal diameter of the maximum transverse diameter(DT)x(DL)and photographic record,intervention in 4weeks after the golden mouse back sebaceous glands spot skin tissue,HE staining to observe the golden ground squirrels,sebaceous glands organization form immune histochemical method to detect PPAR gamma,ABCA1 expression levels.Results:1.Sebaceous patch area of golden hamster: Before intervention,there was no statistical significance in the size of sebaceous patch area of golden hamster among all groups(P > 0.05),which was comparable.With the passage of time,the sebaceous spots of golden hamster in each group increased to different degrees,and the difference was statistically significant(P < 0.05).After intervention,there was no significant difference in the area of sebaceous glands of golden hamster between blank group and pomegranate peel polyphenol cream low-concentration group(P > 0.05).The area of sebaceous glands in rosiglitazone group,positive drug group,pomegranate peel polyphenol cream medium concentration group and high concentration group were lower than that in blank group and pomegranate peel polyphenol low concentration group,with statistical significance(P <0.05).There was no significant difference in the area of sebaceous gland patches among rosiglitazone group,pomegranate peel polyphenol cream medium concentration group,high concentration group and positive drug group(P > 0.05).2.Sebaceous gland spot structure: After intervention,compared with blank group,the volume and number of glandular lobes in each drug group were reduced,and the number of overlapping layers was reduced,and the arrangement was loose,among which the pomegranate skin polyphenol cream medium-concentration group,high-concentration group,positive drug group and rosiglitazone group were more significant.3.Expression of PPARγ and ABCA1: PPARγ expression in pomegranate skin polyphenol cream group,positive drug group and rosiglitazone group at all concentrations were increased compared with blank group(P < 0.05),ABCA1 expression in medium and high concentration groups and positive drug group was increased compared with blank group(P < 0.05),and there was no statistical significance in ABCA1 expression between low concentration group and blank group(P > 0.05).There was no significant difference in the expression of PPARγ and ABCA1 between medium and high concentration groups and positive drug groups(P > 0.05).Conclusions:1.Medium-concentration and high-concentration pomegranate peel polyphenol cream can effectively inhibit the growth of golden hamster sebaceous patches,while low-concentration pomegranate peel polyphenol cream has no significant effect in inhibiting the growth of golden hamster sebaceous patches.2.Pomegranate peel polyphenol cream can improve the structure of sebaceous glands,make the sebaceous gland leaves shrink,reduce the number,loose arrangement,especially in medium and high concentration of pomegranate peel polyphenol cream.3.Pomegranate peel polyphenol cream may regulate sebum metabolism and improve the occurrence and development of acne by activating PPARγ/ABCA1 signaling pathway.Among them,medium and high concentrations of pomegranate peel polyphenol cream can significantly up-regulate the expression levels of PPARγ and ABCA1.
