Based On IGF-1, Explore The Mechanism Of Pomegranate Peel Polyphenol Cream Interfering With Acne Sebum Metabolism

Objective: The purpose of this experiment is to investigate whether pomegranate peel polyphenol cream can inhibit sebaceous gland enlargement and sebum secretion by measuring the size of sebaceous gland spots and observing the sebaceous gland tissue structure,and to further clarify whether pomegranate peel polyphenol cream can further affect sebum metabolism by adjusting IGF-1 and determine the optimal concentration through immunohistochemical detection of IGF-1 expression in sebaceous gland spots.Methods: Thirty-six male golden hamsters,weighing 120± 10 g,were randomly divided into six groups after adaptive feeding for 10 days:blank group,matrix group,low concentration pomegranate peel polyphenol cream group,medium concentration pomegranate peel polyphenol cream group,high concentration pomegranate peel polyphenol cream group,and positive drug group(tretinoin cream),which were labeled with staining method.Before the experiment,each golden hamster was weighed and recorded,and the hair on the clean back was removed with an electronic hair shaver to fully expose sebaceous glands on both sides.The maximum transverse diameter(DT)and maximum longitudinal diameter(DL)of bilateral plaques were measured with vernier caliper,and recorded.The blank group did not interfere.The experimental group(matrix,low,medium and high concentration groups)were coated with matrix cream,low,medium and high concentration pomegranate peel polyphenol cream respectively.The positive drug group was coated with tretinoin cream.Each golden hamster was coated with about1 g cream on both sides of sebaceous gland spots twice daily for4 weeks.After the experiment was completed,anesthetized animals were weighed,back hairs were removed again with a shaver,the size of sebaceous glands on both sides were measured with verniercalipers,photographs were taken and data were recorded,the skin of sebaceous glands on the back of golden hamsters was cut,the size was about 1*1cm,immediately placed in a centrifuge tube filled with 4% formaldehyde fixing solution for fixation,and the marks were made for examination.HE staining was used to observe the histopathological structure of sebaceous plaques,and immunohistochemical method was used to detect the expression of IGF-1 in sebaceous glands.Results: 1.Size of sebaceous gland spot:there are significant differences between groups.compared with the blank group,there are significant differences in low concentration group,medium concentration group,high concentration group and positive drug group(P<0.05).Compared with the low concentration group,the medium concentration group,the high concentration group and the positive drug group have significant differences(P<0.05).There was no significant difference between the middle concentration group,high concentration group and positive drug group(P>0.05).2.Tissue structure of sebaceous gland spot:there are significant differences between groups in sebaceous gland thickness.compared with the blank group,there are significant differences between the medium concentration group and the positive drug group(P<0.05),but there is no significant difference between the medium concentration group and the positive drug group(P>0.05).There were significant differences in the degree of looseness of sebaceous glands among the groups.compared with the blank group,there were significant differences in the medium concentration group,high concentration group and positive drugs(P<0.05),but there were no significant differences among the medium concentration group,high concentration group and positive drugs(P>0.05).3.Expression of IGF-1: there were significant differences among the groups.compared with the blank group,there were significant differences in low concentration group,medium concentration group and highconcentration group(P<0.05).Compared with the low concentration group,there were significant differences between the medium concentration group and the high concentration group(P<0.05).There was no significant difference between medium concentration group and high concentration group(P>0.05).Conclusion: 1.Low,medium and high concentrations of pomegranate peel polyphenol cream can inhibit sebaceous gland growth,and medium and high concentrations of pomegranate peel polyphenol cream can inhibit sebaceous gland spot growth more obviously than low concentrations of pomegranate peel polyphenol cream.2.Medium concentration pomegranate peel polyphenol cream can make sebaceous gland leaf atrophy,number reduction,while medium concentration and high concentration pomegranate peel polyphenol cream can make sebaceous gland loose arrangement.3.Low,medium and high concentrations of pomegranate peel polyphenol cream can inhibit the growth of sebaceous glands and sebum secretion by inhibiting the expression of IGF-1,and the effect of inhibiting the expression of IGF-1 is more obvious in medium and high concentrations of pomegranate peel polyphenol cream.
