Study On The Effect Of Electroacupuncture "Tianshu" And "Neiguan" On Intestinal Motility And D2 Receptors In IBS Model Rats

OjectiveIn this experiment,IBS model rats were used as the research medium to compare the effect of electroacupuncture"Neiguan" and "Tianshu" point on the colon motility of IBS model rats and the related mechanisms from two aspects of "brain" and "intestines".To explore whether there are specific effects in different meridians and acupoints,so as to provide experimental evidence for further studying the specific mechanism of acupoint effect.MethodsSelection of Wistar newborn rats 40 were randomly divided into control group,model control group,Tianshu group and Neiguan group.In the model control group,Tianshu group and Neiguan group,IBS rat model was prepared by mother infant separation and acetic acid enema combined with colorectal expansion.In 9 week old rats were given Tianshu group and Neiguan group electroacupuncture,1 every other day,a total of 5 times.After the intervention,the stool type of IBS rats before and after electroacupuncture was observed by Bristol stool classification.The intestinal sensitivity and contractile intensity were evaluated by abdominal retracement reflex,and the latency and the number of contraction waves were observed.The expression of D2 receptor in DMV and the expression of D2 receptor and c-kit in the colon were detected by immunohistochemistry.Results1.electroacupuncture at "Tianshu","Neiguan" on IBS model rat stool effectBefore acupuncture:compared with the blank control group,the score of stool character of Bristol in model control group,Tianshu group and Neiguan group increased significantly(P<0.01).Compared with the model control group,there was no significant difference between the Tianshu and the Neiguan group(P>0.05).After acupuncture:Compared with the blank control group,the stool score of Bristol in model control group increased significantly(P<0.01);Compared with the model group,Tianshu group and "Neiguan group"group stool scores were significantly reduced(P<0.01);Compared with the pre acupuncture,the scores of Bristol fecal traits in Tianshu group were significantly decreased(P<0.01),and the Bristol fecal score in Neiguan group was decreased(p<0.05).2.Electroacupuncture at "Tianshu","Neiguan" on IBS rat model of abdominal withdraw reflection effectCompared with the blank control group,the latency of the rats in the model control group was significantly shortened and the number of the contraction waves increased significantly(P<0.01),and the latent period of the Neiguan group shortened(P<0.05);Compared with the model control group,the latency of the rats in Tianshu group and Neiguan group was significantly prolonged,and the number of contractile waves decreased significantly(P<0.01).3.Electroacupuncture at "Tianshu","Neiguan" of IBS rats "brain" and "gut" two aspects of intestinal motility factor influence.1)In the dorsum of the vagus nerve:Compared with the blank control group,the positive expression of D2 receptor reactivity in DMV was significantly decreased(P<0.01)in the model control group(P<0.01),and the positive expression of D2 receptor reactivity in the Neiguan group was decreased(P<0.05).2)In the colon:Compared with the blank control group,the positive expression level of D2 receptor immunoreactivity in the colon of the model control group was significantly decreased(P<0.01)and the positive expression level of c-kit immunoreactive substance was significantly increased(P<0.01).The positive expression level of D2 receptor immunoreactivity in the colon of the Neiguan group was decreased,and there was significant difference(P<0.05)and the positive expression level of c-kit immunoreactive substance(P<0.01).Compared with the model control group,the positive expression level of D2 receptor immunoreactivity in the colon of the Tianshu group increased significantly(P<0.01),and the positive expression level of D2 receptor immunoreactivity in the colon of the Neiguan group increased(P<0.05).C-kit immunoreactant Tianshu group and Neiguan group colon expression levels were significantly lower(P<0.01).Compared with group D2 receptor immunoreactive Tianshu,the expression level of rat colon in Neiguan group decreased significantly(P<0.01).Conclusion1 electro acupuncture at "Tianshu" and "Neiguan" can inhibit intestinal contraction,improve intestinal sensitivity and alleviate IBS symptoms in IBS rats."Improved Tianshu"acupoint on gastrointestinal motility disorder obviously,reduce diarrhea effect is stronger.2 the partial mechanism of the Electroacupuncture of "Tianshu" and "Neiguan" on the intestinal motility disorder in IBS rats may be related to the regulation of the D2 receptor in the brain and the content of the D2 receptor and c-kit in the intestine,and the regulation of the two points is different.
