Ischemic heart disease is a kind of disease of high incidence,high morbidity,high mortality and complications,which has become a serious public health and social problem.According to the epidemiological studies,cardiovascular disease is one of the world’s leading chronic diseases.The pathogenesis of ischemic heart disease is mainly about atherosclerosis caused stenosis which results from elevated blood lipids and deposition on the inner walls of arteries as well as myocardial ischemia and hypoxia caused by the thrombosis.Shenloushuangxinwan(SLSXW)derived from the experienced prescription of professor Wang Jie named Shenloushuangxinfang,which consists of Salvia miltiorrhiza,Fructus Trichosanthis,Allium macrostemon,Ginkgo biloba and so on.SLSXW could tonify Qi,recover the blood,invigorate the circulation of blood and dissipating phlegm.And it is intended for the prevention and treatment of stable angina(syndrome of intermin-gled phlegm and blood stasis),which could relieve the symptoms and reduce the recurrence rate of angina.This research and development were supported by the Ministry of Science major new drug projects.In order to provide the evidence,the design and method of this research are based on the functions of SLSXW from the aspects of improving heart function,anti-myocardial ischemic injury,reducing blood lipids,inhibiting thrombosis and improving the blood rheology.The four parts of this research are as follows.1.In order to identify the most suitable preparation technology,we extracted the effective components of SLSXW by four kinds of preparation technology and then the model rat of myocardial ischemia injury which was established by coronary arterial ligating were consecutively gavaged with four kinds of SLSXW.We took the parameters of Ejection Fraction(EF)measured by ultrasonic cardiography as the outcome.2.To observe the myocardial protection effect of SLSXW at different doses,we used the myocardial ischemia-reperfusion injury model.3.To observe the regulation effect of blood lipid of SLSXW at different doses,we used the hyperlipidemia model.4.To observe the influence of SLSXW at different doses on the process of thrombosis and blood viscosity,we used the hyperlipidemia model.1.Influence of SLSXW of four kinds of preparation technology to intervention on heart function with acute myocardial ischemia rat.Objective:To identify the most suitable preparation technology of SLSXW which is used for the treatment of stable angina(syndrome of intermin-gled phlegm and blood stasis)Method:All rats were consecutively gavaged with SLSXW for 5 days consecutively.The model rat of myocardial ischemia injury was established by coronary arterial ligating.Left anterior descending branch of coronary artery was ligated.Rats were consecutively gavaged with SLSXW until the end of the experiment.We took the parameters of Ejection Fraction measured by ultrasonic cardiography as the outcome at 8d and 22d after the operation.Result:The value of EF of model blank group were obviously lower than that of sham operation group at 8d and 22d after the operation.And the value of EF of four experimental groups were improved to varying degrees.The value of EF of group of preparation technology No.1 and No.4 were obviously higher than that of the model blank group.Because of removal the toxic component of preparation technology No.4,we finally choose the preparation technology No.4.Conclusion:Identify the preparation technology No.4 as the most suitable one.2.Pharmacodynamics study of SLSXW_the myocardial protection effect on myocardial ischemia-reperfusion injury modelObjective:To observe the myocardial protection effect of SLSXW on myocardial ischemia-reperfusion injuryMethod:We used the myocardial ischemia-reperfusion injury model and observe the myocardial protection effect by the parameters of serum myocardial enzyme and myocardial infarction range.Result:SLSXW at doses of 0.70g/kg,1.40g/kg,2.8g/kg could obviously reduce the value of Creatine Kinase and Lactate Dehydrogenase 180 min after the operation;SLSXW at doses of 0.70g/kg and 1.40g/kg could obviously reduce the value of Creatine Kinase Isoenzyme 180 min after the operation,while the trend of decreasing Creatine Kinase Isoenzyme by SLSXW at doses of 2.8 g/kg was not obvious.SLSXW at doses of 1.40g/kg and 2.8g/kg could obviously reduce the myocardial infarction range 180 min after the operation.Conclusion:SLSXW have the myocardial protection effect on myocardial ischemia-reperfusion injury by reducing serum myocardial enzyme and myocardial infarction range.3.Pharmacodynamics study of SLSXW_the regulation effect of blood lipid of SLSXW on the hyperlipidemia model.Objective:To observe the regulation effect of blood lipid of SLSXW on the hyperlipidemia model.Method:We used the hyperlipidemia model and observe the regulation effect of SLSXW from different dosage by the parameters of blood lipid.Result:SLSXW at doses of 0.70g/kg could obviously reduce the value of total cholesterol 4w after the operation;SLSXW at doses of 0.70g/kg and 1.40g/kg and 2.80g/kg could obviously reduce the value of triglyceride;SLSXW at doses of 0.70g/kg and 1.40g/kg had the trend to reduce the value of low-density lipoprotein;SLSXW at doses of 0.70g/kg and 1.40g/kg and 2.80g/kg could not improve the value of high-density lipoprotein obviously.SLSXW at doses of 2.80g/kg could obviously reduce the value of total cholesterol 8w after the operation;SLSXW at doses of 0.70g/kg and 1.40g/kg and 2.80g/kg could obviously reduce the value of triglyceride and low-density lipoprotein;SLSXW at doses of 1.40g/kg and 2.80g/kg could improve the value of high-density lipoprotein obviously.Conclusion:SLSXW have the effect on decreasing the value of total cholesterol,triglyceride and low-density lipoprotein and increasing the value of high-density lipoprotein.4.Pharmacodynamics study of SLSXW_the influence of SLSXW on the process of thrombosis and blood viscosity of acute blood stasis ratsObjective:To observe the influence of SLSXW at different doses on the process of thrombosis and blood viscosityMethod:We used the acute blood stasis model which was established by epinephrine hydrochloride injected subcutaneously combined with ice-water bath.The outcomes included thrombosis length and blood viscosity.Result:SLSXW at doses of 1.40g/kg and 2.80g/kg could obviously shorten the length of thrombosis and decrease the wet and dry weight of the thrombosis;SLSXW at doses of 2.80g/kg could obviously induce the blood viscosity on condition of 60S-1 and 5S-1 shear rate.Conclusion:SLSXW have the effect on shortening the length of thrombosis and decreasing the wet and dry weight of the thrombosis and inducing the blood viscosity,which proves that it could improve the condition of acute blood stasis.To sum up,SLSXW is a kind of Chinese herbal compound which is used for the treatment of stable angina(syndrome of intermin-gled phlegm and blood stasis)and has the function of improving cardiac function,anti-myocardial ischemia,anti-hyperlipidemia,inhibiting thrombosis and reducing the viscosity of whole blood,which provides pharmacological base for the further research.
