BackgroundBioinformatics has become an important tool for clinical related scientific research.The GSMD family is the key regulator of pyroptosis.We used bioinformatics methods to explore the expression level and related effects of GSDMD in the occurrence and development of GBM.We found that in the open tumor database,compared with non-tumor brain tissues,the expression of GSDMD was up-regulated in gliomas,and the expression level of GSDMD was significantly correlated with tumor grade,IDH1 mutation status and overall survival.In further in vitro experiments,the relationship between GSDMD and the cell activity of glioma cell lines verified this result.Therefore,we believe that GSDMD is a new biomarker for predicting the prognosis of gliomas.ObjectiveTo investigate the effect of GSDMD gene expression on glioma.MethodsBioinformatics method was used to analyze the glioma gene sequencing information in TCGA database.The differential genes between glioma lesions and adjacent tissues were screened and functional cluster analysis was carried out to understand the potential mechanism of glioma pathogenesis.Select the differential gene of interest(GSDMD),check its expression in all kinds of tumors in TCGA tumor database,and further analyze its expression characteristics in glioma patients in TCGA database,and whether it has an impact on the prognosis of glioma patients.Then,the glioma cell lines with high expression of GSDMD were selected for GSDMD gene knockout.Through cell proliferation,migration,apoptosis and other experiments to explore whether GSDMD is closely related to the occurrence and development of glioma,in order to verify the analysis results of gene information of glioma patients in TCGA database.Results1.TCGA database,there are 3317 differential genes(more than 10 times difference)between glioma and paracancerous tissues,including 1171 up-regulated differential genes and 2146 down-regulated differential genes.2.GSDMD plays an important role in inflammation,and there is a significant difference between glioma and paracancerous tissue.Preliminary analysis of GSDMD showed that in TCGA database,the expression of GSDMD decreased in 8 common tumors,increased in 12 common tumors,and had no significant difference in 13 common tumors.3.GSDMD is highly expressed in gliomas,and the elevated level is significantly correlated with prognosis.4.GSDMD in different grades of gliomas.In the same grade of gliomas,the prognosis of patients with high GSDMD was worse.5.After the silencing of GSDMD gene in the cell experiment,the cell activity and migration ability decreased significantly,at the same time,the apoptosis increased and the level of scorch death-related protein decreased.Conclusion1.The GSDMD of glioma is significantly higher than that of normal tissue.2.The expression level of GSDMD increased with the increase of malignant degree of gliomas,and the increase of GSDMD level in different grades of gliomas was negatively correlated with prognosis.3.In the process of occurrence and development of glioma,GSDMD does not exist as an independent biomarker to judge the prognosis,and the increase of its expression level is closely related to the pathway of pyroptosis.4.Inhibition of GSDMD can inhibit the proliferation and migration of glioma cells and promote their apoptosis. |
