Objective:1. To analysis the expression of monocyte chemoattractant protein-1(MCP-1),tu-mor-associated macrophages(TAMs) in different grades of human glioma paraff-in tissue biopsy specimens, and so we understand relationship between MCP-1, TAMs and pathological grade of glioma.2. To explore the role of MCP-1/CCR2axis in proliferation and migration of glioma, so we can provide experimental basis for the in-depth study of the ro-le of MCP-1/CCR2axis in the related biology behavior of glioma.Method:1. To detect the expression rate of MCP-1, CD68in different grades of human glioma paraffin tissue biopsy specimens with the immunohistochemistry(SABC). The experiments in vitro:1) With MTT assay, to detect the effect of MCP-1(10-160ng/ml), specific re-ceptor antagonist RS504393(5-80ng/ml) for CCR2in certain concentration ra-nge to the proliferation of C6glioma cells;2) With scratch test, to detect the effect of MCP-1(40ng/ml),RS504393(20n g/ml) to the migration of C6glioma cells.Results: 1. In glioma tissue of different grade,the positive expression rate of MCP-1,CD68, which were present brownish yellow in the membrane and cytoplasm of cell were significantly different(p<0.05), the higher the pathological grade of glioma, the more obvious level the positive expression rate of MCP-1,CD68(p<0.05);The experiments in vitro:1) The receptor of CCR2is expressed in the membrane and cytoplasm of C6glioma cells;2) under the influence of MCP-1,RS504393, C6glioma cells are significantly intervened by them.Conclusion:1. There is a significant positive correlation between the expression of MCP-1, CD68and clinical pathological grade of glioma;2. The promoted role of MCP-1/CCR2axis in proliferation and migration of C6glioma cells is significant; specific receptor antagonistRS504393for CCR2c-an significantly inhibit the proliferation and migration of C6glioma cells;3. Further explore the mechanism of the MCP-1/CCR2axis will provide a cer-tain degree of experimental basis for understanding related biology behavior of glioma.
