Objective: This study intends to investigate prognostic signifcance of IL-35 expression in human gliomas,finding new therapeutic targets for gliomas.Methods: Tumor specimens from 77 patients with glioma(neither chemotherapy or radiotherapy before surgery)and 15 normal brain tissue with brain trauma who underwent intracranial decompression surgery were collected from a tertiary hospital from 2019 to 2022.According to the grading system developed by the World Health Organization(WHO),classifies gliomas from I(lowest malignant degree,best prognosis)to IV(highest malignant degree,worst prognosis),of which the low level Glioma(13 cases in WHO I class,25 cases in WHO II class),high-grade glioma(16cases in WHO III class,23 cases in WHO IV class).In this study,immunohistochemistry and western blotting were used to detect the expression of p35 and EBi3 in normal brain tissues removed by intracranial decompression in patients with gliomas of different grades and patients with brain trauma.The follow-up survey was used to calculate the correlation between the survival cycle of different grades of glioma patients and the expression of p35 and EBi3 in the tissues.SPSS24.0 software was used for processing.The measurement data was expressed as `x ±s.Chi-square test was used to investigate the differences of p35 and EBi3 expression in normal brain tissue and different grades of gliomas.The survival curve was analyzed by Kaplan-Meir method.Results: The strong positive expression rate of p35 and EBi3 in human glioma were significantly higher than that of the control group(P <0.05),and the strong positive expression rate of p35 and EBi3 in the high-grade glioma group were significantly higher than that of the low-grade group(P <0.05).Long-term follow-up investigation showed that patients with high IL-35 expression had shorter survival periods than patients with low IL-35 expression,and the difference was statistically significant(P <0.05).Conclusion: In human glioma,the higher the expression of IL-35,the higher the degree of malignancy of glioma,and the shorter the patient’s survival period,IL-35 may act as an indicator to evaluate the malignant degree of glioma and the prognosis of glioma patients.
