Correlation Between Cerebrospinal Fluid Soluble Platelet-derived Growth Factor Receptor β And Cognitive Damage And Cerebrospinal Fluid Biomarkers In Alzheimer’s Disease

Objective:To investigate the relationship between the concentration of soluble platelet-derived growth factor receptorβ(s PDGFRβ)in the cerebrospinal fluid(CSF)of patients with Alzheimer’s disease(AD)and the degree of cognitive impairment and CSF biomarkers.Methods:50 patients with"Probable AD"and 33 patients with normal cognitive function in the Department of Neurology of Anhui provincial hospital were included in this study from September 2018 to August 2020.The neuropsychological evaluation was conducted.When the score of Clinical Dementia Rating scale(CDR)≤1,the patients were considered as mild AD group.Otherwise,the moderate to severe AD group is included.General information and cognitive function of the three groups were compared.Then,CSF s PDGFRβ,CSF amyloid-β(Aβ)1-40,CSF Aβ1-42,CSF total tau protein(T-tau),CSF phosphorylated tau protein(P-tau)were determined by enzyme-linked immunosorbent assay(ELISA)among groups.If Apolipoprotein E(?)4(Apo E(?)4)gene was carried,patients with AD were considered as Apo E4+group,otherwise put them into Apo E4group.Among the three groups the level of CSFs PDGFRβwere compared,and the correlation between it and the cognitive scores and the contents of CSF Aβ1-42,Aβ1-42/Aβ1-40,T-tau and P-tau were studied.Results:(1)It was matched in age,gender and educational level among mild AD group,moderate to severe AD group and control group,and the proportion of hypertension/ diabetes is similar(all P>0.05).The control group performed best in all the cognitive assessments and got highest scores in the Mini mental state examination(MMSE)as well as the Montreal Cognitive Assessment scale(Mo CA)(all P<0.001),while the scores of Activity of Daily Living Scale(ADL)and CDR were significantly lower than those in mild AD group and the moderate to severe AD group(all P<0.001).Compar-rison of CSF biomarkers among the three groups showed that CSF Aβ1-42 and Aβ1-42/Aβ1-40 in mild AD group and moderate to severe AD group were lower than those in control group(P<0.01),while CSF T-tau and P-tau increased in mild AD group and moderate to severe AD group(all P<0.001),but there was no significant difference in CSF Aβ1-40 among the three groups(P>0.05).(2)Analysis of variance showed that there were differences in the concentration of CSF s PDGFRβamong control group(184.88±52.94 pg/ml),mild AD group(219.30±69.71pg/ml)and moderate to severe AD group(235.36±86.19 pg/ml)(F=3.901,P=0.024).It was higher in the mild AD group and the moderate to severe AD group than in the control group by post hoc test(t=2.002,2.613,P=0.049,0.011).(3)There was no significant difference between the Apo E4+group(219.49±76.75pg/ml)and the Apo E4-group(222.80±81.67 pg/ml)(t=-0.127,P=0.900).(4)The level of CSF s PDGFRβwas positively correlated with CSF P-tau(r=0.425,P=0.019)in the mild AD group,but not with Aβ1-42,T-tau,MMSE or Mo CA scores,neither in the control group nor in the moderate to severe AD group(all P>0.05).Conclusions:Expression of s PDGFRβin CSF of AD patients is increased,and may be related to P-tau.
