Generalized tonic-clonic seizures(GTCS)is one of the common types of generalized epilepsy,characterized by loss of consciousness and systemic symmetrical convulsions.Numerous previous studies have shown abnormal functional connectivity(FC)involving extensive regions of the cerebral cortex and cerebellum in patients with GTCS.In recent years,a great of previous studies based on static functional connectivity analysis have found that the FC characteristics between cerebral cortices and the cerebellum,but there are few researches focus on dynamic functional connections.Consequently,the study based on dynamic functional connectivity of cerebello-cerebrum to investigate the temporal and spatial variability,and the state-specific connectivity pattern and the dynamic functional state interactions of dynamic FC in cerebrum were analyzed by K-means clustering.In order to research temporal and spatial variability of cerebro-cerebellar of high-order dynamic functional connections,the study collected resting state f MRI data from 102 patients with GTCS and 106 healthy controls.And the study selected AAL and BN_atlas templates as region of interest and two sliding window methods.The two sliding window methods used in this research got similar results.Under two kinds of sliding window methods and two kinds of templates,for cerebrum,compared with healthy subjects,the research has found significantly increased temporal variability of angular gyrus and decreased temporal variability of occipital-mid and occipital-inf in patients with GTCS,for cerebellum,the temporal variability of cerebellar Crus II was lower in patients with GTCS than in healthy controls.In terms of spatial variability,the DMN-VAN of connectivity was significantly increased,connectivity of VN-VN was decreased,and connectivity of bilateral cerebellum-9 was increased in patients with GTCS compared with healthy controls.Abnormal connection between DMN and VAN may lead to dysfunction in the functioning of task positive network and task negative network,which may be associated with abnormal cognitive function in patients with GTCS.Decreased connectivity variability of the cerebellar Crus II and increased connectivity variability of bilateral cerebellum-9 may result in motor imbalance and motor learning disability,which may be associated with motor function impairment in GTCS patients.To further explore cerebrum of connection in the specific states and dynamic functional state interaction of patients with GTCS,the study adopted K-means clustering to evaluate cerebrum’s dynamic sliding windows and obtained four states.It is found that there was no significant difference in fraction time and mean dwell time between patients with GTCS and healthy controls.For the connection in the four states,state1 is almost negative connection,state2 is positive connection between primary networks and negative connection between superior networks and primary network,state3’s connection is close to state4,but the connection in state4 is almost positive.In the dynamic functional state interaction analysis,the research found that the increased similarity between state4 and state1,decreased similarity between state4 and state2,the finding indicates that the information flow transmission of GTCS patients to the cerebrum is disordered.In general,this study used the combination of high-order correlation and dynamic functional connectivity to research the variability of GTCS,and explored the brain area variability under the brain-cerebellar interaction and the interaction between brain states.The study found high variability within DMN and significant changes between DMN and other positive networks,suggesting that it is related to cognitive impairment in patients with GTCS.At the same time,the interaction between states indicates abnormal information flow in patients with GTCS,leading to disorder of states,which provides evidence for further explaining the abnormal discharge of patients with GTCS.
