Traditional Chinese medicine has a long history and is also an important part of traditional medicine in the world.The diversity of chemical components of traditional Chinese medicine determines the diversity of its pharmacological effects and clinical efficacy,and also leads to some traditional Chinese medicines with varying degrees of toxicity.Traditional Chinese medicine processing is an important part of clinical medicine in traditional Chinese medicine,and its main purpose is to"reduce toxicity and increase efficiency".How to exert the best clinical effect of traditional Chinese medicine after processing has always been the subject of many scholars.As a traditional Chinese medicine,Polygala Radix has the functions of calming the nerves,nourishing the mind,removing phlegm,and reducing swelling.It also has certain gastrointestinal toxicity.Therefore,its processed products are generally used in clinical medicine to achieve the effect of"attenuating toxicity and increasing efficiency"and maximize its medicinal value.There are many processing methods of Polygala Radix,and currently it is mainly processed by auxiliary materials.Among them,Polygala Radix processed by glycyrrhiza is a processed product with many clinical applications,which is included in the Chinese Pharmacopoeia,but its specific processing parameters are not clear.In order to standardize the processing technology of Polygala Radix and ensure the effectiveness of clinical medication,it is necessary to use modern scientific and technological means to explore the best processing technology of Polygala Radix processed by glycyrrhiza,and provide scientific data support and guidance for the optimization of its processing technology.Therefore,in this paper,Polygala Radix processed by glycyrrhiza is selected as the research object,and the L9(3~4)orthogonal test method combined with the quantitative analysis of ultra-high performance liquid chromatography-triple quadrupole-tandem mass spectrometry(UPLC-MS/MS)is used to optimize the processing technology of Polygala Radix,and from the level of cells and animals,the optimal processing technology was verified from the dual perspectives of reducing toxicity and synergy,and finally determined the best processing technology of Polygala Radix processed by glycyrrhiza.The main research contents are as follows:1.Study on processing technology and content determination of Polygala Radix processed by glycyrrhizaUsing the L9(3~4)orthogonal test method,setting the amount of licorice,the amount of water used for decocting and the frying power as the investigation factors,a total of 27 batches of samples were prepared.Combined with UPLC-MS/MS technology and Waters Mass Lynx 4.1 data processing system,The effects of different processing technology on the content changes of 7 kinds of index components in Polygala Radix were classified,quantitatively analyzed,and orthogonal verification experiments were carried out,and the data was statistically analyzed by SPSS software.Finally,the best processing technology of Polygala Radix processed by glycyrrhiza was selected as A2B2C3:12 g of licorice,add an appropriate amount of water,decoct twice,remove slag,combine the filtrate and concentrate to 40m L,add100g Polygala Radix,simmer for 30min,fry under 300W power until the soup is exhausted,take out,and dry it.2.Based on Aβ25-35-induced PC12 cell injury model to evaluate the in vitro activity of different processed products of Polygala RadixAβ25-35-induced PC12 cell injury model was constructed,and an Alzheimer disease(AD)in vitro activity evaluation model of Polygala Radix was established from the cellular level.To observe the protective effect of Polygala Radix and its different processed products on the injury of PC12 nerve cells caused by Aβ25-35,and to carry out statistical analysis combined with the results of the orthogonal test,to preliminarily verify the scientificity of the determination of the optimal processing technology from the perspective of enhancing efficacy.The results showed that under the condition of A2B2C3,the protective effect of Polygala Radix was the most obvious,and the protective effect of its processed product 1 was the weakest.Combined with the results of statistical analysis,it was determined that A2B2C3was the best processing technology.3.Pharmacodynamics evaluation of Polygala Radix processed by glycyrrhiza in the treatment of Alzheimer’s diseaseIn this chapter,on the basis of the evaluation results at the cell level,the pharmacodynamics evaluation of Polygala Radix and its processed products 5 and 1under different processing techniques were carried out.A rat model of Alzheimer’s disease(AD)was constructed,and the Morris water maze behavioral experiment was used to investigate its learning and memory ability and histopathology by HE staining,immunohistochemistry and biochemical index determination.The results showed that the P5 group had the best effect,followed by the P1 group and the raw Polygala Radix group.4.Toxicity evaluation of different processed products of Polygala RadixReducing toxicity is one of the most important indicators to be investigated in the processing of toxic traditional Chinese medicines.Therefore,this chapter uses the mouse acute toxicity experiment and gastrointestinal toxicity experiment to analyze the raw Polygala Radix and its processed products 5 and 1 under different processing techniques were carried out to explore the changes of toxicity before and after processing of Polygala Radix,and to preliminarily verify the safety and scientificity of the optimal processing technology from the perspective of reducing toxicity.The results showed that Polygala Radix had relatively strong acute toxicity and gastrointestinal toxicity,and the toxicity was reduced after processing.Among them,the P5 toxicity is the lowest.To sum up,in this paper,the change of the content of the index components of Polygala Radix is used as the inspection index,and the optimal processing technology of Polygala Radix is determined.The rationality,feasibility and scientificity of the selected processing technology were further verified from the dual perspectives of toxicity reduction and efficiency enhancement,thus providing scientific data support for the optimization of the processing technology of Polygala Radix.
