To Explore The Pharmacological Mechanism Of "Guben Huayu Tongluo Recipe" In The Treatment Of Diabetic Kidney Disease Based On Network Pharmacology

ObjectiveThis study used network pharmacology method to explore the effective active ingredients and pharmacological mechanism of Guben Huayu Tongluo Recipe in the treatment of diabetic kidney disease(DKD).MethodsThe effective active ingredients corresponding to Guben Huayu Tonglu Recipe were collected through TCMSP and BATMAN-TCM database,and the active ingredient structures were input into the Swiss Target Prediction database to obtain the corresponding related targets of the active ingredients."Diabetic kidne y disease、diabetic nephropathy " were input into Genecards database to obtain the related targets of DKD.The above targets were input into Venny online mapping tool to get the intersection targets,and the potential intersection targets of Guben Huayu Tonglu Recipe for the treatment of DKD were finally obtained.Then the String database and Gephi software were used to construct protein-protein interaction network(PPI),and the Cytoscape software was used to construct the network of "herbs-active ingredients-potential targets",and the core ingrediens and core targets of Guben Huayu Tongluo Recipe for the treatment of DKD were screened out according to the degree of value.Then,the GO biological function analysis and K EGG metabolic pathway analysis of potential targets were carried out by DAVID database.According to the number of targets involved,important signaling pathways of Guben Huayu Tongluo Recipe for the treatment of DKD were screened out.Finally,C ytoscape software was used to construct the network of "core ingredients-core targets-important signaling pathways" of Guben Huayu Tongluo Recipe in the treatment of DKD.Results1.Guben Huayu Tongluo Recipe for the treatment of DKD involves 90 active ingredients,and the core ingredients include quercetin,kaempferol,isorhamnetin,hesperidin,palmidin A,etc.2.Guben Huayu Tongluo Recipe plays a therapeutic role in DKD by acting on384 potential targets,and the core targets involve AKT1,MAPK1,VEGFA,STAT3,PIK3 CA,IL6,PIK3R1,MAPK3,etc.3.Guben Huayu Tongluo Recipe in the treatment of DK D involves 965 biological processes such as cell proliferation and apoptosis,regulation of inflammatory response,112 cell components such as cytoplasm and nucleus,and 188 molecular functions such as protein binding and enzyme binding.4.Guben Huayu Tongluo Recipe for the treatment of DKD by regulating 142 metabolic pathways,mainly involving PI3K-Akt signaling pathway,RAS signaling pathway,MAPK signaling pathway,HIF-1 signaling pathway,mTOR and AMPK signaling pathways,etc.Conclusion1.Flavonoids such as quercetin,kaempferol,isorhamnetin,hesperidin,as well as palmidin A and Folsaeure are the main active ingredients of Guben Huayu Tongluo Recipe for the treatment of DKD.2.Guben Huayu Tongluo Recipe for the treatment of DK D mainly by acting on targets such as AK T1,MAPK1,VEGFA,STAT3,PIK3 CA,IL6,PIK3R1 and MAPK3,and by regulating multiple signaling pathways such as PI3K-Akt signaling pathway,RAS signaling pathway,MAPK signaling pathway,HIF-1 signaling pathway,mTOR and AMPK signaling pathways.
