Study On The Mechanism Of Fuzheng Huayu Jiangzhuo Tongluo Recipe In The Treatment Of Chronic Renal Failure Based On Network Pharmacology

ObjectiveThrough the method of network pharmacology,this paper studies the effective ingredients,targets and signal pathways of Fuzheng Huayu Jiangzhuo Tongluo Recipe in the treatment of chronic renal failure,and intends to clarify its mechanism of action,so as to provide reference for later basic research.MethodsThrough TCMSP,TCMID,BATMAN-TCM database and literature mining,the active ingredients of Fuzheng Huayu Jiangzhuo Tongluo Recipe were collected,Pub Chem and Swiss ADME database were used to verify the molecular structure of the active ingredients,Pharmmapper and Uni Prot database were used to predict and correct the target of active ingredients,Genecards and OMIM database were used to construct the target database of chronic renal failure;The intersection of Fuzheng Huayu Jiangzhuo Tongluo Recipe and chronic renal failure target library was obtained by Venny online mapping tool,and the key targets were obtained by analyzing and screening the intersection targets through STRING database,David database was used to complete GO enrichment analysis and KEGG pathway analysis of key targets;Gephi and Cytoscape software should be used for analysis of the result.Results1.Fuzheng Huayu Jiangzhuo Tongluo Recipe in the treatment of chronic renal failure involves 316 active ingredients,such as Mairin,Emodin-1-O-beta-D-glucopyranoside,Physciondiglucoside,Ederagenin,Torachrysone-8-O-beta-D-(6’-oxayl)-glucoside,Bifendate,etc;2.Fuzheng Huayu Jiangzhuo Tongluo Recipe in the treatment of chronic renal failure involves 130 key targets,such as PIK3 CA,AKT1,MAPK3,EGFR,VEGFA,etc;3.Fuzheng Huayu Jiangzhuo Tongluo Recipe in the treatment of chronic renal failure involves 358 biological processes,such as negative regulation of apoptotic process,positive regulation of cell proliferation,extracellular exosome,extracellular space,etc;4.Fuzheng Huayu Jiangzhuo Tongluo Recipe in the treatment of chronic renal failure involves 96 signal pathways,such as PI3K/Akt,HIF-1,VEGF,TGF-β,Toll-like,NF-κB,Leukocyte transendothelial migration,B cell receptor,T cell receptor,etc.Conclusion1.Anthraquinones and flavonoids may be the key ingredients of Fuzheng Huayu Jiangzhuo Tongluo Recipe in the treatment of chronic renal failure.2.Fuzheng Huayu Jiangzhuo Tongluo Recipe may delay renal fibrosis by regulating the expression of PI3K/Akt,HIF-1,VEGF,TGF-βand other pathways,reducing apoptosis and inhibiting the synthesis of extracellular matrix.3.Fuzheng Huayu Jiangzhuo Tongluo Recipe may play a protective role in kidney by regulating the expression of Toll-like,NF-κB,Leukocyte transendothelial migration,B cell receptor,T cell receptor and other signal pathways.
