Objective: Based on network pharmacology and experimental analysis of quercetin on oral lichen planus immune intervention research,through the analysis of network pharmacology results,as well as in vitro study of quercetin on oral lichen planus in patients with peripheral blood lymphocyte proliferation,apoptosis,migration and IL-6,IFN-γ cytokine secretion level changes,in order to provide reference for further study of quercetin on the treatment of oral lichen planus related mechanism.Methods: The drug targets of quercetin were searched and screened by traditional Chinese medicine system pharmacology database and analysis platform(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP),and the corresponding genes of the targets were searched by Uniprot database.The possible mechanism of quercetin on diseases was predicted by Kyoto Encyclopedia of Genes and Genomes(Kyoto Encyclopedia of Genes and Genomes,KEGG)and Gene Ontology(Gene Ontology,GO)enrichment analysis,and the drug-disease-pathway-target network diagram was established by Cytoscape3.8.0.According to the results of network pharmacology analysis,15 patients with patchy oral lichen planus,15 patients with erosive oral lichen planus and 15 healthy volunteers were selected.Peripheral blood T lymphocytes were extracted and isolated.CCK8,flow cytometry and cell migration assay were used to detect the effects of quercetin on lymphocyte proliferation,apoptosis and migration.The secretion levels of IL-6 and IFN-γ cytokines were detected by ELISA.Results: A total of 75 quercetin-related targets and 6110 differential genes in oral lichen planus disease were screened by TCMSP,and 66 differential genes corresponding to common targets were screened out,and a total of 116 signaling pathways were enriched,among which the key pathways were AGE-RAGE signaling pathway,IL-17 signaling pathway and HIF-1 signaling pathway.Enriched to 1474 biological processes,the key biological processes are reactive oxygen species metabolism,reactive oxygen species stimulation reaction,cytochemical stress reaction,etc.In the basic experimental study,through the different results of the effects of different concentrations of quercetin on cells,it can be seen that when the concentration is 10 μM,quercetin has a significant proliferation and migration effect on peripheral blood T lymphocytes in patients with oral lichen planus.When the concentration is increased to 40 μM,quercetin has a significant effect on the apoptosis of T lymphocytes.For the results of enzyme-linked reaction adsorption assay,it can be seen that quercetin inhibits the secretion of IL-6 in peripheral blood T lymphocytes and promotes the secretion of IFN-γ with the increase of quercetin concentration.Conclusion: Comprehensive network pharmacology and in vitro studies have shown that quercetin may interfere with the occurrence and development of oral lichen planus by regulating the balance of Th1 and Th2 cells in peripheral blood of OLP patients and affecting the secretion of related cytokines.
