Objective:To investigate the expression and significance of IL-35signaling pathway related factors in oral lichen planus.Methods:1.40 cases of patients with oral lichen planus peripheral blood(20 cases of non-erosive type,20cases of erosive type)and 20 cases of healthy control group.The m RNA levels of Ebi3,p35,gp130,IL-12Rβ2,STAT1 and STAT4 in peripheral blood mononuclear cells(PBMCs)of OLP were measured by real-time fluorescence quantitative PCR(qPCR).The above factors and OLP were analyzed.The condition and the correlation of immune indicators.2.Oral lichen planus lesions in 41 cases(21 non-erosive type,20 cases of erosive type)and 15 healthy control groups.The expression of Ebi3,p35,gp130,IL-12Rβ2,STAT1,STAT4 mRNA in OLP lesions was detected by real-time fluorescent quantitative PCR(qPCR).Proteins Ebi3,p35,and gp130 in OLP lesions were detected by immunohistochemistry.The distribution and expression of IL-12Rβ2,and the relationship between these factors and the clinical characteristics of OLP.Results:1.The expression levels of Ebi3,p35,gp130,IL-12Rβ2,STAT1 and STAT4 mRNA in peripheral blood of oral lichen planus were higher than those in normal control group(P<0.05);STAT4 was higher in erosive than non-erosive type,and the rest There was no statistical difference between the two types of OLP(P>0.05).The correlation between the above factors and OLP immune parameters was analyzed.The results showed that the expression level of Ebi3 mRNA was related to T+B+NK cells(rs=0.349,P=0.040),and the expression level of STAT1 mRNA was positively correlated with IgA(rs=0.468,P=0.003),the expression level of STAT4mRNA was negatively correlated with Ig M(rs=-0.334,P=0.038).2.The expression levels of Ebi3,p35,gp130,IL-12Rβ2,STAT1 and STAT4 in oral lesions of oral lichen planus were higher than those in normal control group(P<0.05);Ebi3,p35,gp130,IL-12Rβ2 in OLP lesion tissues.The positive expression rates ofprotein were 66%(27/41),39%(16/41),36.59%(15/41),and 34.15%(14/41),respectively,which were higher than those in the normal control group.Significance(P<0.05);and the relative expression of Ebi3 and p35 protein was positively correlated(rs=0.404,P=0.009).The positive expression rate of Ebi3 protein was higher in the basal cell liquefaction group than in the low group(c2=9.172,P=0.010);the positive expression rate of IL-12p35 protein was higher in the erosive type than in the non-erosive type(c2=7.220),P=0.007).The positive expression rate of IL-35 protein in OLP lesions was 34%(14/41),which was higher than that of the normal control group.The expression of IL-35 protein in erosive type was higher than that of non-erosive type(c2=4.364,P=0.037).The difference was statistically significant(P<0.05).The expression of gp130 and IL-12Rβ2 protein in erosive OLP lesions was significantly higher than that in non-erosive ones(c12=5.707,P1=0.017;c22=4.264,P2=0.037),and the difference was statistically significant(P<0.05).The relationship between IL-35 ligands and receptor expression levels was further analyzed.The results showed that IL-35 receptors and There was a positive correlation between the expression levels of ligands(rs=0.892,P<0.001).The positive expression rate of gp130 and IL-12Rβ2 protein in the OLP group had no significant differences in the course of disease,lymphocyte infiltration,and basal cell liquefaction(P>0.05).Conclusion:1.The expression of IL-35 signaling pathway related factors in peripheral blood of oral lichen planus is increased,which is related to immunity,suggesting that IL-35signaling pathway related factors may be involved in the pathogenesis of OLP immunity.2.The expression of IL-35 and its receptors gp130 and IL-12Rβ2 was up-regulated in OLP lesions,which was related to the clinical classification of OLP,suggesting that IL-35 signaling pathway related factors may be involved in the formation and development of OLP lesions.
