Objective:To investigate the inhibitory effect of Baihe Gujin decoction on non-small cell lung cancer in vitro and in vivo.To explore the biological mechanism of Baihe Gujin decoction in the treatment of non-small cell lung cancer.Methods:1.First of all,the effect of Baihe Gujin decoction on cell activity was detected by CCK-8 experiment,and the effect of Baihe Gujin decoction on the proliferation of non-small cell lung cancer cells was detected by cell clone formation test and Ed U,so as to explore the tumor inhibitory effect of Baihe Gujin decoction on non-small cell lung cancer in vitro.2.The model of subcutaneous transplanted tumor was established on the back of nude mice and divided into control group and experimental group treated with Baihe Gujin decoction.Four weeks later,the nude mice were killed and the tumor was removed.The tumor size between the two groups was compared and statistically analyzed.The expression of tumor proliferation and apoptosis related genes was detected by immunohistochemistry to explore the tumor inhibitory effect of Baihe Gujin decoction on non-small cell lung cancer in vivo.3.Finally,the anti-cancer mechanism of Lily Gujin decoction was explored by flow cytometry and Western blotting.Results:1.With the increase of the concentration of Baihe Gujin decoction,the activity,proliferation and clone formation ability of NSCLC cells decreased significantly.2.After oral administration of Baihe Gujin decoction for 30 days,the growth rate of tumor volume slowed down and the volume of the tumor was smaller than that of the control group,which was statistically significant(P<0.05).After the tumor was fixed,the paraffin-embedded sections were stained by immunohistochemical method,and it was found that the expression of Ki67 was significantly down-regulated and the expression of,Cleaved caspase-3 was significantly increased in the experimental group.3.The results of flow cytometry showed that NSCLC cells in the experimental group had obvious G0G1 phase arrest and obvious apoptosis,which inhibited cell proliferation.The results of Western Blot suggested that the expression of apoptosis-related proteins Bad,Bax and Cleaved cas increased with the increase of drug concentration under the action of different concentrations of BHGJT,on the contrary,the expression of apoptosis-related protein Bcl-2 decreased in a concentration-dependent manner.Conclusion:1.Baihe Gujin decoction can inhibit the activity,proliferation and clone formation of NSCLC in vitro.2.Baihe Gujin decoction can induce apoptosis and inhibit proliferation of NSCLC cells in vivo.3.Baihe Gujin decoction can inhibit cancer by inducing G0G1 arrest and apoptosis of NSCLC cell cycle.
