Background:The aim of this study was to confirm whether osteonecrosis of the femoral head(ONFH)has a certain relationship with the infection of the pathogen by means of sequencing.Methods:The patients with ONFH were selected from Liaocheng People’s Hospital,and the inclusion criteria were clinically definite for idiopathic patients without any obvious risk factors.The patient’s 27 information was extracted from the hospital’s medical record information system.Fresh necrotic femoral head tissue specimens were collected within half an hour in vitro,and bite into pieces of bone tissue as small as possible with a rongeur.After pulverizing treatment femoral head necrosis,the eluted DNA/RNA solution is amplified and nucleic acid is constructed according to standard methods.The metagenomic next-generation sequencing(m NGS)was performed with the BGISEQ-50 gene sequencer(Huada Gene Company),and the microbial nucleic acid sequence in the sample was analyzed based on high-throughput sequencing technology.After the results are obtained,and the pathogenicity data is analyzed in combination with pathogens to determine the pathogenicity of the pathogen.Results:A totele of 6 patients of idiopathic ONFH were obtained in the study.All patients showed similar clinical manifestations,such as limb gait,reduced Harri scores and X-ray imaging features.The 38 m NGS results of necrotic femoral head tissue showed some specimens detected few sequence of Pseudomonas aeruginosa,and other detected microbes also showed few fragments.Simultaneously,suspected background microorganisms were detected in specimen sequencing,such as Ralstonia insidiosa,Propionibacterium acnes,Burkholderia ubonensis,etc.No virus,fungus,parasites,M.tuberculosis complex and mycoplasma/chlamydia were detected.Conclusions:Although we have not found specific ONFH-related pathogens in necrotic femoral head tissue,our research are an area that has never been studied before which may be provides some theoretical help for future research in the future.
