Objective To study the change of proliferation and apoptosis of malignant melanoma A375 cells that was intervented Qinghai red lycium barbarum polysaccharide of a certain concentration Method A375 cells were cultured in vitro experimental model.Using red lyciun barbarun polysaccharide power prepared as different concentration of drug culture mediun to intervente A375 cells,and set control group(treated with nomal culture mediun) at the same time.Then using CCK-8 assay detected absorbance values of each group of cells and calculate the corresponding proliferation inhibition rate.Flow cytometer was used to detect the apoptotic rate with Annexin V-PI staining and the cellular mitochondrial membrane potential with JC-1 staining. Result CCK-8 assay experiment indicated that cells proliferation inhibition rate in each dose group was gradually increased with the increasing of concentration of lycium barbarun polysaccharide,the differences were statistically significant(P<0.05),that is to say the Qinghai red lycium barbarum polysaccharide could inhibit the proliferation of A375 cells,and it is dependent on concentrations.Annexin V-PI staining experiments showed that apoptotic rate of each dose group was different(P<0.05),but it is not dependent on concentrations;JC-1 staining experiment showed that the ratio of cells in the experimental group had decreased mitochondrial membrane potential was higher than that in control group(P<0.05).These two sets of experiments showed that Qinghai red lycium barbarum polysaccharide could induce apoptosis of A375 cells. Conclusion Qinghai red lycium barbarum polysaccharide in a certain concentration could inhibit the proliferation of malignant melanoma A375 cells and promote its apoptosis. |
