Objectives The quaternary ammonium salt monomer of 2-Methacryloxylethyldo decylmethyl ammonium bromide(2-Methacryloxylethyldodecylmethyl ammonium bromide,MAE-DB)was processed and synthesized.By detecting the cytotoxicity and antibacterial properties of MAE-DB,better root canal irrigation agents and root canal sealers were provided in clinic,and better technical support was provided to improve the success rate of root canal retreatment.Methods The experiment was divided into three parts: 1.Synthesis of MAE-DB:After the intermediate product was obtained by using N-methyldiethanolamine and bromododecane as raw materials,methacryloyl chloride was added to react to generate the required experimental drug MAE-DB.The molecular weight was determined by mass spectrometer,which proved to be the required experimental drug.2.Detection of the effect of MAE-DB on cell proliferation: MAE-DB was dissolved in deionized water to prepare 5μg/m L,10μg/m L,15μg/m L,20μg/m L,25μg/m L,30μg/m L,35μg/m L,40μg/m L MAE-DB solution.Mouse fibroblasts(L929)were cultured to the fifth generation,and the prepared MAE-DB solution was added to the cell culture medium that had been adhering to the wall.Each group was set up five vice holes.CCK-8 detection method was used to detect the proliferation rate of L929 cells 6h,12 h,24h and 48 h after adding the drug solution.The safety of MAE-DB was explored and statistically analyzed.3.Antibacterial experiment of MAE-DB: After screening the best antibacterial concentration of the above experimental groups,compared with the commonly used clinical flushing agents such as sodium hypochlorite(sodium hypochlorite,Na Cl O),compoundchlorhexidine(compound chlorhexidine,CHX)and ethylenediaminetetraacetic acid(ethylenediamine tetraacetic acid,EDTA),added to the biofilm of Enterococcus faecalis and Candida albicans that have been cultured for 48 h,and continued to be cultured for 24 h,the activity of Enterococcus faecalis and Candida albicans biofilm was detected by MTT method,and the antibacterial properties of different concentrations of MAE-DB solution,sodium hypochlorite,compound chlorhexidine and ethylenediaminetetraacetic acid were indirectly compared,and the experimental results were statistically analyzed.The live bacteria/dead bacteria fluorescence staining of Enterococcus faecalis and Candida albicans after treatment was carried out.The strength of red fluorescence(dead bacteria)and green fluorescence(live bacteria)was observed by fluorescence microscope.The overlap position of live bacteria and dead bacteria was light yellow or orange.The antibacterial properties of each group were compared and analyzed.Results 1.In this experiment,MAE-DB was synthesized by Menshut gold method and verified by mass spectrometry.2.There was no significant difference in the absorbance(optical density value)between the MAE-DB solution of the experimental group with different concentrations and the control group at the same time point(P>0.05).With the change of time,the OD value of each experimental group was statistically significant(P<0.05).3.Inhibitory effect of MAE-DB on Enterococcus faecalis and Candida albicans biofilm: 8 groups of MAE-DB solution with different concentrations were statistically significant(P<0.05),the best concentration of Enterococcus faecalis biofilm was30μg/m L MAE-DB solution group,the best concentration of Candida albicans biofilm was35μg/m L MAE-DB solution group.The best concentration of the two groups were compared with 3% Na Cl O,CHX group and 17% EDTA group,respectively,with statistical significance(P<0.05).The best effect was MAE-DB solution group.Conclusion 1.MAE-DB solution has no toxic effect on L929 cells and can be considered as a new type of flushing agent.2.MAE-DB solution had inhibitory effect on Enterococcus faecalis and Candida albicans biofilm,which was better than 3%sodium hypochlorite and compound chlorhexidine commonly used in clinical practice.Figure 12;Table 14;Reference 145...
