Objectives To observe the effects of Shuangqingpinghua Recipe on Lee’s index,blood sugar and the expression of skeletal muscle m TOR,Beclin-1 and LC3 protein in rats with metabolic syndrome,and to explore the mechanism of the formula for improving glucose and lipid metabolism.Methods The model of metabolic syndrome was induced by high-fat diet supplemented with 10% fructose water and combined with N-nitro L-arginine methyl ester and streptozotocin.They were randomly divided into model group(MSD group),metformin group(MF group),Shuangqingpinghua prescription high-dose(SQPHF-H),medium-dose(SQPHF-M),low-dose(SQPHF-L)groups and normal group(NC group).The body weight of rats were measured regularly before and after the experiment,Lee’s index was calculated,blood glucose and blood pressure were measured.Cholesterol(TC)and triglyceride(TG)were detected.To observe the establishment of the rat model of metabolic syndrome and the changes of the value of Shuangqingpinghua formula after drug intervention.The pathological changes of skeletal muscle were observed by hematoxylineosin(HE)staining.The morphological changes of skeletal muscle fibers were observed by Masson staining.The average optical density of LC3 protein in skeletal muscle tissue of each group was detected by immunohistochemistry(IHC).The protein expression levels of m TOR,Beclin-1 and LC3 in skeletal muscle tissues were detected by Western blot.Results 1 Compared with the normal group,the body weight of rats in the model group decreased,the amount of water and food intake increased,the amount of urine increased,the stool was soft or thin,the body hair was disorderly,the color was withered,the spirit was listless,and the movement was lazy;In the normal group,rats gained weight,had normal intake of food,water and urine,hard stool,smooth and white body hair,and active spirit.Compared with model group,the above conditions were improved in metformin group and Shuangqingpinghua formula groups.2.Fasting blood glucose(FBG),2h postprandial blood glucose(PG2h),TC and TG in model group were significantly increased(P < 0.01).Compared with model group,serum FBG and PG2 h in Shuangqingpinghua formula group were significantly decreased,and insulin was significantly increased(P < 0.01).Compared with the normal group,the blood pressure,TC and TG of the model group were significantly increased;compared with the model group,the TC and TG of the metformin group and each dose of Shuangqingpinghua formula were significantly decreased(P < 0.01).3 skeletal muscle HE staining showed model group skeletal muscle morphology,quantity and distribution of abnormalities,prompt model group rats skeletal muscle cell damaged and dysfunction,mason dyeing in connective tissue hyperplasia,collagen fibers increase gradually and muscle fiber spacing widened by collagen fiber hyperplasia,immunohistochemical luminosity values significantly lower than other groups,LC3 autophagy proteins in the membrane adhesion is less,Autophagy was inhibited.He staining of metformin group and Shuangqingpinghua formula low-dose,medium-dose and high-dose groups showed that the distribution of skeletal muscle cells was looser and the nucleus had inward shift than that of the normal group.According to the results of Masson staining,the presence of collagen fibers was slightly more than that of the normal group.4.The expression of m TOR protein in skeletal muscle tissue was significantly up-regulated(P < 0.01),and Beclin-1 and LC3 proteins were significantly down-regulated(P < 0.01).Conclusions 1 Shuangqingpinghua formula can down-regulate the blood glucose and lipid levels of MS rats,enhance the tolerance of rats to glucose load,improve the disorder of glucose and lipid metabolism,and correct the abnormal glucose tolerance.2Shuangqingpinghua formula can improve the tissue morphology of skeletal muscle.3Shuangqingpinghua formula can improve the oxidative stress level of skeletal muscle in MS rats.4 Shuangqingpinghua formula can improve insulin resistance in rats with metabolic syndrome,and its mechanism may be related to the activation of m TOR,Beclin-1 and LC3 protein expressions in the autophagy pathway of skeletal muscle tissue.Figure 7;Table 6;Reference 146.
