Objectives To observe the effects of Suaeda salsa on blood sugar,blood lipids,liver tissue pathological morphology and oxidative stress indicators of malondialdehyde,superoxide dismutase,and glutathione peroxidase in type 2 diabetic rats;Explore the effect of Suaeda salsa on the expression of PTP1 B,p-IRS-1,PI3 K,p-AKT,GLUT4 protein in liver tissue of type 2 diabetic rats.Methods High-fat diet combined with intraperitoneal injection of streptozotocin was used to establish a rat model of type 2 diabetes.The 70 SD rats purchased were fed for 1 week,and 10 rats were randomly selected as the normal group and fed with a normal maintenance diet.The remaining rats were fed as the experimental group with high-fat diet.After 4 weeks,rats in the experimental group were fasted with STZ buffer(30mg/kg)for12 hours,and the normal group was given the same dose of normal saline intraperitoneally.The rats were closely observed for 72 hours after the injection.After 72 hours,the modeled rats were subjected to oral glucose tolerance test,with FBG > 11.1 mmol/L or PG2 h >16.7 mmol/L as the modeling standard.The modeled rats were randomly divided into 5groups.The metformin group was given 90 mg/kg per day by gavage,the low-dose Suaeda salsa group was given 405 mg/kg crude drug per day,and the medium-dose Suaeda salsa group was given crude drug per day.The dose of 810 mg/kg was intragastrically administered.The high-dose Suaeda salsa group was intragastrically administered at the crude drug dose of 1620 mg/kg per day.The normal group and the model group were administered the same volume of normal saline.After the rats were administered,the fasting blood glucose was tested every 2 weeks,and the glucose tolerance was tested every4 weeks.After the administration,blood was collected from the abdominal aorta,and the serum was centrifuged to determine glycosylated serum protein,glycosylated hemoglobin,total cholesterol,triglycerides,low-density lipoprotein cholesterol,high-density lipoprotein cholesterol and C-peptide,calculate insulin resistance index,and detect liver tissue Oxidative stress-related indicators,liver tissue HE staining,immunohistochemistry was used to detect liver tissue p-AKT,GLUT4 protein expression,and Western Blot to detect liver tissue PTP1 B,p-IRS-1,PI3 K,p-AKT,GLUT4 protein levels.Results 1 In the model group,fasting blood glucose,2h postprandial blood glucose,area under the glucose curve,glycosylated hemoglobin,glycosylated serum protein levels and insulin resistance index increased,and C-peptide levels decreased;compared with the model group,rats in the Suaeda salsa group had fasting blood glucose,After a meal,blood glucose,area under the glucose curve,glycosylated hemoglobin,glycosylated serum protein levels and insulin resistance index decreased,and C-peptide levels increased.2 The total cholesterol,triglycerides,and low-density lipoprotein cholesterol levels in the liver tissues of rats in the model group increased,and the high-density lipoprotein cholesterol levels decreased;compared with the model group,the total cholesterol and triglycerides in the liver tissues of the Suaeda salsa group Ester,low-density lipoprotein cholesterol levels decrease,and high-density lipoprotein cholesterol levels increase.3 The level of malondialdehyde in the liver tissue of rats in the model group increased,and the levels of superoxide dismutase and glutathione peroxidase were decreased;compared with the model group,the level of malondialdehyde in the liver tissue of rats in the Suaeda salsa group decreased,The levels of superoxide dismutase and glutathione peroxidase increased.4 HE staining of liver tissue,compared with the model group,Suaeda salsa improved the morphology and structure of liver tissue.5 According to the results of immunohistochemistry,the average optical density of p-AKT and GLUT4 in the model group was significantly reduced.Compared with the model group,the average optical density of p-AKT and GLUT4 in each administration group was significantly increased.Western Blot showed that in the model group,the expression of PTP1 B increased,while the expression of p-IRS-1,PI3 K,p-AKT,and GLUT4 decreased;compared with the model group,Suaeda salsa reduced the expression of PTP1 B increased.The expression level of IRS-1,PI3 K,p-AKT,GLUT4.Conclusions 1 Suaeda salsa can down-regulate the blood glucose level of T2 DM rats,enhance the tolerance of the rat’s OGTT to glucose load,and improve glucose metabolism disorders and impaired glucose tolerance.2 Suaeda salsa can improve blood lipid levels in T2 DM rats.3 Suaeda salsa can improve the oxidative stress level of T2 DM rats.4 Suaeda salsa can improve the tissue morphology of the liver.5 Suaeda salsa can improve blood glucose,blood lipids and oxidative stress levels in T2 DM rats,which may be related to the regulation of PI3K/AKT/GLUT4 signaling pathway.Figure 5;Table 8;Reference 106...
