Objectives To optimize the extraction process of dietary fiber(DF)of suaeda salsa,complete the preparation of DF of high-purity suaeda salsa and detect its physical and chemical properties,and explore its effect on hyperlipidemia rats.Methods DF of suaeda salsa was extracted by enzymolysis.The ratio of material to liquid,the amount of protease added,enzymolysis time,enzymolysis temperature and pH were used as factors to carry out experiments.The best process parameters were optimized by Box-Behnken design with DF extraction rate as response value;DF adsorption capacity of suaeda salsa was determined by nitrite and cholesterol;60 male SD rats were selected and divided into normal group(10 rats)and modeling group(50 rats).The model group was fed with high-fat diet for modeling.After 80 days,blood samples were taken from orbit to detect the level of blood lipids.The rats with significant difference between the content of blood lipids and the normal control group were included in the experiment as successful high-fat model.40 rats with successful modeling were randomly divided into 5 groups(high fat model group,positive control group,suaeda salsa DF high,medium and low intervention groups).During the experiment,except for the normal control group,the other groups were fed with high-fat feed,and were administered by gavage(normal group and model group were given distilled water of the same amount).After 6 weeks of intervention,the rats’ weight,food intake,liver weight,fat weight around epididymis,liver coefficient and serum were measured respectively blood lipid and other indicators.Results 1 The single factor test conditions were as follows: the ratio of material to liquid was 1∶25(g/ml),the amount of papain added was 16%,the time of enzymatic hydrolysis was 3 h,the temperature of enzymatic hydrolysis was 50℃,pH=7;the best extraction technology was as follows: the ratio of material to liquid was 1∶20(g/ml),the amount of papain added was 20%,the time of enzymatic hydrolysis was 4 h,pH=8,and the temperature of enzymatic hydrolysis was 52℃.2 At pH=2 and pH=7,suaeda salsa DF has the adsorption capacity for nitrite and cholesterol.At pH=2,t=2 h,the adsorption capacity for nitrite is the strongest,and at pH=7,t=30 min,the adsorption capacity for cholesterol is the strongest.3 Compared with the high fat model group,the body weight,food consumption,liver weight,fat weight around epididymis and liver coefficient of the intervention group of suaeda salsa DF decreased,with the largest reduction of 26.21%,26.66%,44.72%,57.14% and 39.92%,respectively.In the aspect of blood lipid,TC,TG and LDL-C were all decreased in the high-fat model group and suaeda salsa DF intervention group,with the maximum decrease of 8.76%,87.20% and 60.50%,respectively.Compared with the positive group,TC level of suaeda salsa DF intervention group was similar,TG effect was not significant,but LDL-C value decreased and HDL-C increased significantly(p<0.05).LDL-C decreased 38.16%,HDL-C increased 27.01%.The results showed that the dietary fiber of suaeda salsa could control the body weight,food intake,liver weight and fat weight around epididymis of hyperlipidemia rats to a certain extent,and it also had the effect of reducing blood fat.Conclusions The extraction process of suaeda salsa DF was optimized,and the preparation of high purity suaeda salsa DF was completed.It was found that it has certain adsorption capacity for NO2-and TC in vitro;It was found that suaeda salsa DF could control the body weight,food intake and the weight of fat around epididymis of hyperlipidemia rats to a certain extent,and could reduce the blood lipid of hyperlipidemia rats.Figure12;Table17;Reference 116...
