Objective Data mining was used to summarize the commonly used Chinese herbal medicines for cough variant asthma,analyze the drug use pattern of cough variant asthma,and summarize the key drug groups for the treatment of cough variant asthma.With the help of network pharmacology,the mechanism of action of the key drug groups will be explored to provide theoretical support for clinical disease treatment and later experimental validation.Methods 1 The literature related to the treatment of cough variant asthma using Chinese herbal medicines published in Chinese databases CNKI,VIP,Wan Fang and CBM since the establishment of the database until December 31,2020 was screened.Screening was performed according to the nadir criteria,and the Chinese herbal medicines of the screened literature were summarized.The high-frequency herbal medicines for the treatment of this disease were identified according to their frequency of use.Association rules and clustering rules were applied to identify the intrinsic associations of high-frequency herbal medicines for the treatment of cough variant asthma,and the key drug groups for the treatment of cough variant asthma were screened.2 The OMIM,Gene Cards,Mala Cards and Dis Ge NET disease databases were searched to obtain the disease genes of cough variant asthma,the active ingredients and corresponding targets of the drugs were obtained through TCMSP and chemical databases,and the drug target names were normalized with the help of Uniprot database.PPI network analysis of protein targets was performed using the STRING database.Data visualization and to pology analysis were performed with Cytoscape V3.7.0 software to screen the key protein targets for the treatment of cough variant asthma.GO function and KEGG pathway enrichment analysis of key protein targets with the help of R language.Results 1 According to the search term "cough variant asthma",a total of 2328 articles were retrieved from CNKI,2943 articles from Wan Fang,1643 articles from VIP,and 1351 articles from CBM.According to the inclusion and exclusion criteria,a total of 1368 articles were obtained,and 321 kinds of Chinese medicines were used in the treatment of cough variant asthma,with a cumulative frequency of 15785 times,among which 26 kinds of single Chinese medicines were used more than 200 times.2 The medicinal properties of Chinese medicine are mostly warm,bitter,pungent and sweet,and the treatment is mainly for the lung,while taking into account the liver,spleen and stomach,and Chinese medicine is mostly used to resolve phlegm,stop cough and calm asthma.3 Based on the results of the cluster analysis,the high frequency drugs can be grouped into 6 categories.The first group includes Hedysarum Multijugum Maxim and Atractylodes Macrocephala Koidz and Saposhnikoviae Radix,The second group includes Citrus Reticulata and Poria Cocos Wolf and Stemonae Radix and Schizonepetae Herba and Platycodon Grandiforus and licorice,The third group includes Scutellariae Radix and Mori Cortex,The fourth group includes Ephedra Herb and Amygdalus Communis Vas and Cicadae Periostracum and Earthworm and White batryticated silkworm and Perillae Fructus and Peucedani Radix and Eriobotryae Folium,The fifth group includes Schisandrae Chinensis Fructus and Asari Radix Et Rhizoma and Paeoniae Radix Alba and Arum Ternatum Thunb,The sixth group includes Asteris Radix Et Rhizoma and Farfarae Flos and Belamcandae Rhizome.4 After correlation analysis,it was found that Ephedra Herb and Asari Radix Et Rhizoma,Ephedra Herb and licorice and Amygdalus Communis Vas,Ephedra Herb and Amygdalus Communis Vas and Cicadae Periostracum and Earthworm were the drug groups with the highest rule support among the correlation terms.5 Combining the results of frequency of drug use and association analysis,Ephedra Herb and Amygdalus Communis Vas and Cicadae Periostracum and Earthworm were the key drug group for this study.6 The possible active ingredients in the key drug group for the treatment of cough variant asthma are quercetin,apigenin,oleic acid,luteolin,stigmasterol,kaempferol.The possible main targets for the treatment of cough variant asthma are TP53,JUN,MAPK1,MAPK3,AKT1,RELA.7 The potential targets of key drug groups in the treatment of diseases may be involved in a variety of biological processes,such as regulating cell response to oxidative stress,lipopolysaccharide,antibiotics and so on.They have the molecular function of binding with cytokine receptors,ubiquitin like protein ligase and DNA binding transcription factors.They may be involved in regulating AGE-RAGE signaling pathway,TNF signaling pathway,IL-17 signaling pathway,PI3 K Akt signaling pathway and C-terminal signaling pathway There are many signaling pathways,such as lectin receptor signaling pathway.Conclusion1 The treatment of cough variant asthma in Chinese medicine is mostly based on the lung theory,and cough suppressing is the basis of medicine for this disease.2 The results of association analysis and cluster analysis were closely related to the classical prescriptions such as Yupingfeng powder,Zhisou powder and Xiaoqinglong decoction,showing the advantages of the classical prescriptions in the treatment of this disease.The new formula(Ephedra Herb and Amygdalus Communis Vas and Cicadae Periostracum and Earthworm and White batryticated silkworm and Perillae Fructus and Peucedani Radix and Eriobotryae Folium)and the key drug group(Ephedra Herb and Amygdalus Communis Vas and Cicadae Periostracum and Earthworm)can be used to regulate the liver and lung internally and externally,providing guidance for clinical selection of drugs.3 The key drug group(Ephedra Herb and Amygdalus Communis Vas and Cicadae Periostracum and Earthworm)for the treatment of cough variant asthma may act through the active ingredients quercetin,apigenin,oleic acid,luteolin,stigmasterol and kaempferol on TP53,JUN,MAPK1,MAPK3,AKT1,RELA target proteins,and regulate AGE-RAGE signaling pathway,TNF signaling pathway,IL-17 signaling pathway,PI3K-Akt signaling pathway,C-type lectin receptor signaling pathway and other signaling pathways,to attenuate airway inflammatory cell infiltration and reduce airway hyperresponsiveness for the purpose of treating cough variant asthma.To attenuate airway inflammatory cell infiltration and reduce airway hyperresponsiveness for the purpose of treating cough variant asthma.
