Objective:This study collected the literature about the treatment of acute pancreatitis by Chinese medicine in recent 10 years,analyzed the rule of Chinese medicine in the treatment of acute pancreatitis,screened the core combination drugs with the highest level of Customs connection,and studied the mechanism of its treatment with network pharmacology technology,which provided the thinking for the clinical prescription of Chinese medicine for the treatment of acute pancreatitis.Methods:Objective to search and collect the TCM prescriptions for the treatment of acute pancreatitis from CNKI and Wanfang database in recent 10 years.First of all,through the statistical module of traditional Chinese Medicine Inheritance computing platform V3.0,the frequency statistical analysis of the nature and taste,meridian tropism and medication category of all prescriptions is carried out.In the prescription analysis module,the frequency statistics,cluster analysis and association rule analysis of traditional Chinese medicine are carried out to obtain the core combination drugs.Secondly,tcmsp database was used to obtain the effective components and traditional Chinese medicine targets of core combination drugs,and genecards database was used to obtain the disease targets and the intersection targets of traditional Chinese medicine and disease.The network control chart of "core combination drug active ingredient intersection target disease" was constructed by using Cytoscape.Finally,the PPI protein interaction network of the intersecting target genes was analyzed by using string database,and the go function and KEGG pathway of the intersecting target genes were analyzed by using R program clusterprofiler data package.Results:(1)A total of 246 literatures were included,including 256 prescriptions.(2)A total of 143 kinds of traditional Chines Abstracte medicine were involved,and the cumulative frequency of use was 2820 times.Rhubarb,Bupleurum bupleurum,Radix Scutellariae,Mirabilite,Citrus aurantii,Magnolia officinalis,Radix Paeoniae paeoniae,Radix yanhuso,Radix Amylostae and Salvia miltiorrhiza were used more than 70 times.Among them,rhubarb was used most frequently with a proportion of96.09%.The top seven drugs in terms of frequency were heat-clearing drugs,purging drugs,qi regulating drugs,blood circulation promoting and blood stasis drugs removing,drugs for deficiency tonifying,drugs surface relieving and drugs dampening.(3)The total frequency of drug four qi was 2583 times,including cold drug flavor1646 times,warm drug flavor 680 times,flat drug flavor 223 times,cool drug flavor23 times and hot drug flavor 11 times.The total frequency of five flavors of drugs was4779 times,including 2327 times for bitter drugs,1185 times for acerbity drugs,562 times for sour drugs,481 times for sweet drugs and 224 times for salty drugs.The total frequency of drug channelization was 8804 times,among which the highest frequency of splenic channelization was 1670 times.(4)According to clustering analysis,can be divided into rhubarb acid-insoluble ash-the drugs,rhubarb,radix scutellariae,radix bupleuri-magnolia bark-mirabilite-magnolia officinalis,radix paeoniae alba,radix scutellariae radix bupleuri-mirabilite acid-insoluble ash-mirabilite-rhubarb-magnolia bark-radix bupleuri-gardenia-white paeony root,rhubarb,radix bupleuri,radix paeoniae alba,glauber’s salt-radix scutellariae,rhizoma corydalis,a total of 4 class.(5)The core drug combination with the highest correlation degree was screened out as rhubarb-Bubuurum-Scutellaria radix-Glauber’s saltpetri-Fructus aurantii-Magnolia officinalis.(6)There were 54 active components in the core combination,and the main chemical components that might be effective for acute pancreatitis included quercetin,kaemol,β-phytosterol,and baicalin,etc.,and 142 effective targets for the treatment of acute pancreatitis might include Akt1,IL6,VEGFA,MAPK3,Casp3,Jun,EGF,MAPK1,MAPK8,EGFR,etc.(7)The treatment of acute pancreatitis with core combination drugs may involve in 2194 biological processes such as cytochemical stress reaction,lipopolysaccharide reaction and oxidative stress reaction;153 molecular functions such as cytokine activity,phosphatase binding and heme binding;58 cell components such as membrane raft,cell capsule cavity,plasma membrane cave like invagination and protein kinase complex;and diabetic complications There are 162 pathways including AGE-RAGE signaling pathway,tumor necrosis factor signaling pathway,interleukin-17,HIF-1 signaling pathway and PI3K / Akt signaling pathway.Conclusion:(1)The location of acute pancreatitis mainly lies in the spleen,which is closely related to the liver,gallbladder,stomach,large intestine,lung and other viscera.At the same time,clearing away heat and toxin,dredging the interior,regulating qi and leading stagnation,promoting blood circulation and removing blood stasis are the treatment principles.(2)The core drug combination(rhubarb Scutellaria baicalensis Radix Bupleurum mirabilite Fructus aurantii Magnoliae officinalis)in the treatment of acute pancreatitis may be through quercetin,kaempferol β-Such factors as AKT1,IL6,VEGFA,MAPK3,CASP3,JUN,EGF,MAPK1,MAPK8 and EGFR play a role in regulating the signaling pathways such as AGE-RAGE signaling pathway,tumor necrosis factor signaling pathway,interleukin signaling pathway,signaling pathway and signaling pathway in diabetic complications.(3)The therapeutic effect of acute pancreatitis focuses on clearing away heat and toxin,dredging internal channels,regulating qi and leading stagnation,promoting blood circulation and removing blood stasis.In modern medical mechanism,it is to prevent and treat pancreatic cell apoptosis,anti-inflammatory and anti-oxidation.Their cognition can provide medication basis for the treatment of AP in the future,and improve the clinical efficacy.
