| Objective:To observe the effect of hepatitis B immunoglobulin(HBIG)and hepatitis B vaccine injected at different time intervals on the proliferation of splenic lymphocyte subsets in mice.To explore the effect of maternal hepatitis B surface antibody(anti-HBs)on humoral and cellular immunity of infants vaccinated with hepatitis B vaccine.Methods:120 BALB/c mice(60 males and 60 females)were fed.According to the time interval between HBIG and the first hepatitis B vaccine,120 mice were randomized into0-week group,2-week group and 4-week group.Then the mice in each group were randomly divided into 1μg subgroup and 2μg subgroup according to the dose of hepatitis B vaccine administrated.Each mouse was injected with 50 IU HBIG at the experiment beginning(0week).The mice in the three groups were injected with the first dose of hepatitis B vaccine at the experiment beginning,in the 2nd and in the 4th week respectively,and then injected with the second and third dose of hepatitis B vaccine every 4 weeks.Spleen lymphocytes were isolated from all mice 4 weeks after the third dose of hepatitis B vaccine.The proportion of lymphocyte subsets(CD3~+,CD3~+CD4~+,CD3~+CD8~+and CD3~-CD19~+)were detected by flow cytometry.Results:The proportion of CD3~-CD19~+B lymphocytes related to humoral immunity in 1μg subgroup and 2μg subgroup was higher in 4-week group than that in the 0 and 2-week group,and the differences were statistically significant(male mice in 1μg subgroup F=7.501,P=0.003;female mice in 1μg group F=14.759,P<0.001;male mice in 2μg group F=13.708,P<0.001;female mice in 2μg group F=13.733,P<0.001);The proportion of T lymphocytes and its subsets(CD3~+CD4~+and CD3~+CD8~+T lymphocytes)related to cellular immunity were not higher in 4-week group than that in the other two groups.Compared 1μg subgroup with 2μg subgroup,the proportion of CD3~-CD19~+B lymphocytes in 2μg subgroup was higher than that in 1μg subgroup in both male and female mice in 4-week group,and the differences were statistically significant(male mice t=-2.740,P=0.014;female mice t=-2.388,P=0.028);The proportion of T lymphocyte subsets related to cellular immunity did not show that it was higher in 2μg group than in 1μg group.The proportion of CD3~-CD19~+B lymphocytes in male mice was higher than that in female mice in the 0 and 2-week group.Conclusion:After injection of HBIG to mice,delayed administration of hepatitis B vaccine and increasing the dose of hepatitis B vaccine contribute to the proliferation of B lymphocytes,but delayed administration of hepatitis B vaccine has no obvious effect on the proportion of T lymphocyte subsets,which suggests that maternal anti-HBs may affect the humoral immunity of infants after hepatitis B vaccination. |
