| OBJECTIVE: In this experiment,a model of liver inoculation with qi stagnation and blood stasis syndrome combined with tumor cells was established in order to explore the effect of this peach kernel-safflower medicine on liver function,inflammatory factors and inflammatory factors after improving the pathological state of qi stagnation and blood stasis in rats Impact on the autologous clearance of tumors.Methods: Clean SD rats were randomly divided into 6 groups: blank control(normal group),simple tumor(model group),qi stagnation and blood stasis + tumor(composite model group),qi stagnation and blood stasis tumor + red Flower(safflower group),Qi stagnation and blood stasis bearing tumor + peach kernel(peach kernel group),Qi stagnation and blood stasis bearing tumor + peach kernel safflower(combined group).The compound model group,safflower group,peach kernel group,and combination group were given chronic unpredictable mild stress(CUMS)for 35 days to establish a model of qi stagnation and blood stasis.Mice in other groups were fed normally.After 35 days,the general conditions of the rats were observed and the opening experiment,pain threshold experiment and sucrose preference test were carried out to verify the qi stagnation and blood stasis model.Then,rat ascites cancer cells(Walker-256)were inoculated into the model group,composite model group,safflower group,peach kernel group and combination group.Among them,the compound model group,safflower group,peach kernel group and combination group are all rat models of Qi stagnation and blood stasis and tumor inoculation.On the 7th day after tumor inoculation,traditional Chinese medicine intervention was started for 2 weeks,and the model group was given distilled water by gavage.After 14 days of continuous administration,blood samples,liver tissues and tumor tissues were collected from rats.Detect cytokines,make pathological sections and immunohistochemical staining.Results: The opening results of rats and the sucrose preference experiment confirmed that chronic stress induction can induce the establishment of a model of qi stagnation and blood stasis in rats.By comparing the liver quality between the groups,it was found that peach kernels and safflower can slow down the growth of tumors after symptomatically improving the symptoms of qi stagnation and blood stasis in rats,and have certain anti-tumor activities.In terms of tumor inoculation,rats in the state of qi stagnation and blood stasis are more likely to inoculate tumors,that is,the compound model has lower body weight than the model group.From the results of liver function,compared with the normal group,each The content of ALT and AST in the group increased significantly(P>0.05).Compared with the model group,the content of the compound model group increased significantly(P>0.05).The results of tumor necrosis factor and vascular endothelial growth factor also support this result.Compared with the normal group,the inflammatory factors in other groups were significantly increased(P>0.05).Compared with the model group,the content of the compound model group increased(P<0.05);in the results of immunohistochemistry,after CD4/CD8 cells were stained,compared with the model group,the cell content in the liver tissue of the compound model was less,indicating that the rat Immunity is reduced,and the ability to auto-clear tumor cells is reduced,which supports the above results.Conclusion: The use of chronic and unpredictable compound stimulation can establish a satisfactory rat model of qi stagnation and blood stasis;the method of direct injection of tumor cells can establish a rat model of liver carcinoma in situ.The modeling time is short and the operation is simple.The tumor growth pattern and inoculation rate are considerable;peach kernel-safflower can improve the stagnation of Qi and blood stasis in tumor-bearing rats,improve liver function,reduce the expression of inflammatory factors;and improve the immunity and tumor clearance rate of rats. |
