Prostate cancer is one of the common malignant tumors in the male urinary system.At present,the conventional treatment of prostate cancer has obvious side effects.It has been reported that active substances isolated from natural plants have certain anti-tumor activity.Anthocyanin is an active substance commonly found in natural plants.It has the effects of inhibiting the production of oxidative free radicals,inhibiting the release of inflammatory factors and inhibiting the growth of tumor cells.The PI3K/Akt signaling pathway plays an important role in the proliferation of prostate cancer cells.Some studies suggest that anthocyanins may be an inhibitor of the PI3K signaling pathway and play an anti-tumor effect by inhibiting the PI3K/Akt signaling pathway.In recent years,literature has pointed out that anthocyanins of Lycium ruthenicum have a variety of biological functions,including anti-aging,antioxidant,vision protection,and prevention of cardiovascular and cerebrovascular diseases.However,there are few reports on the anti-tumor effects and mechanisms of anthocyanins of Lycium ruthenicum Objective Anthocyanin is a natural plant pigment with a variety of biological effects.Related studies have found that anthocyanins have a certain inhibitory effect on the growth and metabolism of tumor cells.Lycium ruthenicum contains a lot of anthocyanins and has strong biological activity.However,it is not clear whether anthocyanins of Lycium ruthenicum can inhibit the growth of tumor cells.The purpose of this study was to investigate the effects and possible biological mechanism of the main anthocyanin monomer(Pt3G)of Lycium ruthenicum Murray on prostate cancer DU-145,LNCaP and PC-3 cells.Provide a new basis for the biological effect of Pt3G to the prevention and treatment of tumor.Methods After intervention of prostate cancer DU-145,LNCaP and PC-3 cells with a concentration of 0-800 μg/mL of Pt3G solution for 24 h,cell proliferation was detected by MTT method,and cell apoptosis was detected by flow cytometry and TUNEL method.Use flow cytometry to detect cell cycle changes,JC-1 staining to detect changes in intracellular mitochondrial membrane potential and mitochondrial membrane permeability,ROS staining to detect changes in intracellular ROS content,and western blot to detect changes in cells The expression levels of PTEN,PI3K,p-PI3K,Akt,p-Akt,Bcl-2,Bax,cytochrome c,caspase 3and other related factors.Results(1)MTT results showed that Pt3G inhibited the proliferation of DU-145,LNCaP and PC-3 cells in a concentration-dependent manner.The IC50 values were 361.58 μg/mL,601.97 μg/mL,and 445.79 μg/mL(p <0.05).Observation through an inverted microscope showed that with the increase of Pt3G concentration,the adherence ability of DU-145,LNCaP and PC-3 cells decreased in a dose-dependent manner,and the cell morphology gradually changed from spindle to round.(2)The results of flow cytometry and TUNEL showed that Pt3G treated DU-145,LNCaP and PC-3 cells for 24 h,the apoptosis rate increased significantly and had a dose-dependent effect(p <0.05).(3)Pt3G can block the cell cycle of DU-145,LNCaP and PC-3 in S phase(p <0.05).(4)The results of mitochondrial JC-1 staining showed that Pt3G can induce the increase of mitochondrial membrane permeability in DU-145,LNCaP and PC-3 cells and release cytochrome c into the cytoplasm(p <0.05).(5)ROS staining results show that Pt3G can induce intracellular ROS production in a concentration-dependent manner(p <0.05).(6)Western bolt results show that Pt3G can significantly down-regulate the expression levels of PI3K,p-PI3K,Akt,p-Akt,Bcl-2 and other factors(p <0.05),and significantly increase Bax,cytochrome c,caspase 3,cleaved The expression level of factors such as caspase 3(p <0.05).Pt3G can achieve the effect of anti-DU-145,LNCaP and PC-3 cell growth and promote the occurrence of cell apoptosis through the ROS/PTEN/PI3K/Akt/caspase 3 pathway.Conclusion Pt3G of Lycium ruthenicum Murray can inhibit the proliferation of prostate cancer DU-145,LNCaP and PC-3 cells by inducing cell apoptosis.Its mechanism of action is related to the regulation of ROS/PTEN/PI3K/Akt/caspase 3 signaling pathway.
