Study On The Structure Of Gut Microbiota In Patients With Psoriasis Based On 16S RRNA Sequencing

Objective: The intestinal flora was compared by the 16 S rRNA gene sequence analysis in the psoriasis patients and the healthy volunteers,to exploring the structural characteristics of intestinal flora in psoriasis patients,and to providing the new direction for the treatment of psoriasis.Methods: The stool specimens were collected from 12 psoriasis patients(group P)and 12 healthy volunteers(group C),followed by for DNA extraction,PCR amplification of target fragments,library construction,and sequencing.The results were analyzed by the OTU cluster analysis,α-diversity analysis,β-diversity analysis,and so on.Results:1.There were no statistically significant differences in gender,age,and BMI between the experimental group and the control group(P>0.05);2.There were 5,430 OTU obtained by the OTU cluster analysis.Among them,2667 OTU were found in two groups,1,596 only in group P,and 1167 only in group C;3.There was no statistical difference in the abundance and diversity of the intestinal flora between the two groups,according to the result of theα-diversity analysis(P>0.05);4.There was significant statistical difference in the structure of the intestinal flora between the two groups,according to the result of theβ-diversity analysis(P<0.05);5.There were significant differences of more than 10 species in the abundance between the two groups by the species diversity analysis.In the group P,the number of Clostridia,Erysipelotrichaceae,Peptostreptococcaceae and Blautia increased,but the number of Negativicutes,Veillonellaceae,Dialister,Megamonas and Ruminococcaceae＿UCG-002 reduced.Conclusions:1.Compared with the healthy control,there was no statistical difference in the abundance of the intestinal flora in the psoriasis patients;2.Compared with the healthy control,there was significant statistical difference in the structure of the intestinal flora in the psoriasis patients;3.The structure of intestinal flora in patients with psoriasis is characteristic.4.Changes in intestinal flora structure may be related to the pathogenesis of psoriasis.
