The steamed chicken soup with Panax notoginseng is a famous winter nourishing medicine diet in Yunnan Province,China.It has been popular among Chinese since the Qianlong period.At present,there are few studies on the steamed chicken soup with Panax notoginseng.Firstly,there is no systematic study on the preparation process;and then the blood-enriching is only popular among the people,and there is no scientific and systematic research on the blood-enriching.These reasons seriously restrict the development of the steamed chicken soup with Panax notoginseng industry.In this study,the components of different the preparation technology in steamed chicken soup with Panax notoginseng were identified and analyzed,after that the preparation technology was optimized based on the blood-enriching,so as to explore the effect of the steamed chicken soup with Panax notoginseng on postpartum anemia,hemorrhagic anemia,myelosuppression anemia and the mechanism of myelosuppression anemia.This study has far-reaching significance for the development and promotion of Panax notoginseng steamed chicken industry.The main results are as follows:1.Establishment of HPLC and analysis of sample differenceHPLC method for determination of steamed chicken soup with Panax notoginseng was established.The recovery of 13 saponins was 100%±RSD was less than 2.7%between 5%;The standard curve R2 was more than 99%;HPLC was established for all samples of Panax notoginseng steam pot chicken.The similarity of13 peaks of Panax notoginseng saponins reached more than 90%.The main components and cluster analysis of saponins in the steam pot of notoginseng were carried out.When the class spacing is 10,12 samples are divided into two categories,S7 and S8 are one and the other is one.The cumulative variance contribution rate of the two main components,Panax Notoginsenoside R1and ginsenoside Rg1,was 86.1%,which could represent most of the information of saponins in the steam pot of Panax notoginseng;The steaming time was 6 hours,the combination of chicken and Panax notoginseng was 20:1,which had a great correlation with main components and good comprehensive quality.The results of HPLC showed that the lower the ratio of chicken to Panax notoginseng,the higher the relative content of saponins in the steam pot of Panax notoginseng;When the ratio of chicken to Panax notoginseng is 20:1,100:1 and200:1,the higher the type and content of transformed saponins is,the smaller the total saponins are,the higher the quality of freeze-dried powder is.Under the condition of the ratio of steaming time to chicken Panax notoginseng,the content of saponin in the residue was>the content of chicken saponin>the content of saponin in chicken soup.The 272 compounds were identified by LC-MS,which can be divided into seven categories:phenolic acid,flavonoids,nitrogen compounds,tannins,lignans and coumarins,terpenes and other categories.The content of triterpenes and flavonoids in chicken and Panax notoginseng was the highest.2.Study on the optimization of the preparation technology of the chicken in the steam pot of Panax notoginsengThe best conditions for promoting the hematopoietic function of mice were:the ratio of the steamed chicken soup with Panax notoginseng was 20:1,and the steaming time was 5.5 hours.3.The improvement effect and mechanism of steamed chicken soup with Panax notoginseng on different anemiaThe results showed that the blood routine and organ index of the mice with postpartum anemia,acute hemorrhagic anemia and myelosuppression anemia were restored to the same state as that of the blank group.The activity of IL-3 and EPO in kidney and serum can be increased,and TNF-αcan be reduced.The activity activated JAK2-STAT5 signaling pathway,promoted the proliferation of hematopoietic cells,and alleviated anemia symptoms caused by cyclophosphamide. |
