Objective:By studying the effect of Shugan Jieyu Capsule on the protein expression and m RNA expression of VIP,c AMP,AQP3 factors in the stomach and colon tissues in a rat model of functional constipation,explore the relevant mechanism of Shugan Jieyu Capsule on functional constipation,And provide an objective basis for its treatment of functional constipation.Methods:SD(Sprague-Dawley)rats were randomly divided into 6 groups with 10 rats in each group.They were functional constipation model group,blank group,Shugan Jieyu capsule high-dose group,Shugan Jieyu capsule medium-dose group,Shugan Jieyu capsule low-dose group,and cisapride capsule group.The blank group did not do any treatment,and the other groups were given the compound diphenoxylate suspension by gavage to establish a functional constipation rat model.Stomach and colon tissues were taken after 4 weeks of treatment.ELISA method was used to detect the expression of VIP,c AMP,AQP3 protein in rat stomach and colon tissue,and RT-q PCR method was used to detect the expression of VIP,c AMP,AQP3 m RNA in rat stomach and colon tissue.Results:After modeling,compared with the blank group,the model group and each treatment group showed longer defecation time for the first black stool and significantly reduced fecal water content,the difference was statistically significant(P<0.05);rats were treated with compound diphenoxylate After 4 weeks of oral administration of the suspension,in addition to the prolonged time of the first black stool and decreased fecal water content,there were also rough hair,withered yellow,dry and hard fecal particles,reduced fecal particles,reduced food intake,and decreased water consumption.Reduced activity,lying in a pile or even curled up in the corner of a squirrel cage,emotional depression,anxiety,and susceptibility to fright,further illustrate the success of the functional constipation model.ELISA and RT-q PCR detection of related factors in rat stomach and colon tissue showed that compared with the blank group,the expression of VIP,c AMP and AQP3 in model rats decreased(P<0.05);after treatment,compared with the model Compared with the group,the expression of VIP,c AMP and AQP3 in each treatment group increased(P<0.05).Among them,the increase of Shugan Jieyu Capsule’s medium and low dose group and the cisapride group was obvious,while the increase of Shugan Jieyu Capsule high-dose group was not obvious.The expression trends of related factors detected by ELISA and RT-q PCR are roughly the same.Conclusions:After modeling,the expression of VIP,c AMP,AQP3 genes and proteins of rats all decreased,the time for the rats to discharge the first melancholy was prolonged,and the water content of feces decreased significantly,and the general condition was poor.After treatment,the expressions of related factors and proteins have all recovered,and they are generally in good condition.It shows that Shugan Jieyu Capsule can reduce the symptoms of functional constipation by increasing the expression of VIP,c AMP,and AQP3 in the stomach and colon of rats with functional constipation,and has a therapeutic effect.
