The Shugan Jieyu Capsule Improved Clinical Outcomes And Decreased Hyper-connected Brain Functional Connectivity Of Mild To Moderate Depression Patients

Objective:To evaluate effects of Shugan Jieyu on mild to moderate depression patients,the depressive symptoms and cognitive functions were assessed by clinical scale and resting-state neuro-imaging.Methods:According to strict inclusion and exclusion criteria,52 patients with mild to moderate depression and 38 normal controls were enrolled.Eight weeks of follow-up studies were performed.Depressive symptoms were assessed by Hamilton Depression Scales at baseline and after the 8th week of treatment.Cognitive function was evaluated by Webster’s Intelligence Test and Wechsler Memory Test.Then the resting-state functional magnetic resonance(RS-fMRI)scan was performed at baseline and after 8 weeks of treatment.Results:Before and after treatment in patients with mild to moderate depression,the Hamilton Depression Scale score was significantly lower(p<0.01),and the mean score was normal at the time of grouping.The number of intelligence quotients and memory quotient in patients with mild to moderate depression was significantly lower than that in the normal control group.The intelligence and memory quotient of patients with mild to moderate depression were significantly improved after treatment.There was no significant difference between the group and the normal control group.FC results showed increased connectivity between right basal ganglia and right frontal gyrus,right basal ganglia and left iliac crest,right basal ganglia and right iliac crest of patients than control(p<0.05).After Shugan Jieyu Capsule treatment,the functional connectivity of these brain regions was significantly lower in patients,and there was no significant difference in the functional connectivity between the group and the normal controls(p>0.05).The simple effect of treatment group and control group was significant.The results were corrected by GRF,p<0.05 after calibration,and cluster size>5 The functional connection between the caudate nucleus and the right frontal cortex was positively correlated with the total IQ change(R=0.288,P<0.05).Conclusion:Shugan Jieyu Capsule can improve symptoms and cognitive function in patients with mild to moderate depression.Before and after treatment,patients with depression have basal ganglia and frontal gyrus and bilateral frontal cortex compared with normal healthy control group changes significantly.The functional connection between the caudate nucleus and the right frontal cortex in the drug intervention group was positively correlated with the total IQ change.To understand the mechanism of Shugan Jieyu Capsule in improving depression and cognitive function in patients with depression,and provide a new basis for clinical treatment of depression.
